- Acting in the Dark: During the auditions, Scott Menville had no idea he was reading for a new incarnation of Doctor Octopus. All information about the show down to its name was secret at the time, with auditioning voice actors receiving fake show titles and character names. Even the descriptions were vague, with Scott only having "a 19 year old high school teacher named Professor Harrison" to go off of.
- Actor Existence Limbo: "Generations" features an appearance from Chameleon, but due to Patton Oswalt being unavailable, he's played by Valenzia Algarin while disguised as Anya's stepsister Maria.
- Acting for Two:
- Josh Keaton voices Norman Osborn, John Jameson, and Ollie Osnick.
- Ben Diskin voices Spencer Smythe and Flash Thompson.
- Jason Spisak voices Alistair Smythe and Scorpion.
- Jim Cummings voices Hammerhead and Ghost.
- To the surprise of no one (despite a rumor otherwise saying John DiMaggio would voice the former character), Fred Tatascoire voices the Hulk in addition to Max Modell, Crossbones, and Beetle.
- Gwen Stacy, Black Widow, and Crimson Dynamo are all voiced by Laura Bailey.
- Yuri Lowenthal, the voice of Clayton/Clash, also voices Curt Connors/The Lizard and Nocturnal.
- Scott Menville voices Otto Octavius and Grady Scraps. It also helps that Grady replaces Otto in Season 3.
- Adored by the Network: It doesn't take up all the airtime, but it does have daily reruns, and episode premieres are rerun a few times on premiere day.
- As of this writing (June 2018), it's the only currently running Marvel Universe cartoon to actually have reruns on weekdays, as opposed to Avengers Assemble and Guardians of the Galaxy mainly being shown only on Sundays, despite the latter two having more episodes.
- All-Star Cast: We have Robbie Daymond as the title character, with major characters played by Laura Bailey, Ben Diskin, Josh Keaton, John DiMaggio, Max Mittelman, Fred Tatasciore, Jason Spisak, and even Patton Oswalt as Uncle Ben of all people. What a crew!
- Approval of God: Josh Keaton expressed support for Robbie Daymond being the new Spider-Man voice.
- Ascended Fanon: A strange example. In this incarnation, Gwen's costumed identity is referred to as Spider-Gwen instead of Spider-Woman or Ghost Spider as in the comics.note Justified in that, this version of Gwen doesn't even bother wearing a mask, being unconcerned with a secret identity.
- Casting Gag: After being well-known as one of Spider-Man's definitive voices, Josh Keaton was cast as Spidey's nemesis Norman Osborn in this series.
- Crossdressing Voices: Hammerhead's son Tully is voiced by Laura Bailey.
- The Danza: Joe the coffee shop owner is voiced by Joe Quesada.
- Dawson Casting: As per usual with all Spider-Man shows, where we have the teenage characters voiced by adult voice actors, even in the case of Nadji Jetter, the voice of Miles Morales, who's the youngest of the show's main voice actors. The only exceptions so far are Sandgirl, who's voiced by Sofia Carson, and Herman Schultz Shocker, who's voiced by Cameron Boyce.
- Descended Creator:
- Joe the coffee shop owner is voiced by Joe Quesada.

- We have American voice actor Troy Baker applying a Russian accent for Kraven The Hunter.

- In a technicality, this applies to Venom. When he made his appearance in the episode "Venom", he was voiced by Ben Diskin due to how, just for that episode only, he took over Flash Thompson, one of his regular characters in the series. In season 2, Venom reappears, having taken over Eddie Brock, who's voiced by the same voice actor as him, Ben Pronsky.
- Production wise, Kevin Burke and Chris Doc Wyatt, who served as consulting producers in season 1, have been promoted to supervising producers in season 2, replacing Kevin Shinick. Likewise, Collette Sunderman takes over as the primary voice director, rather than sharing with Kris Zimmerman Salter like in season 1.
- Mr. Salerno was voiced by Sean Schemmel in season 1 and Phil LaMarr in season 2.
- Playing Against Type:
- Scott Menville is usually known for playing heroes or at least good guys, yet here he's billed to play Doctor Octopus, a villain. It's eventually subverted when be becomes a hero during the second half of season 2, and dies a hero in the season 2 finale.
- We have Josh Keaton, who played Spider-Man in The Spectacular Spider-Man as Norman Osborn.
- Ben Diskin, usually known for playing heroic characters, or at least good guys, plays Spencer Smythe. While he has done villains before, such as Eddie Brock, a.k.a Venom in one of the previous series, they're usually close in age to the protagonist, and not a middle aged man like here.
- Teala Dunn's previous animated roles include Tuck from Wonder Pets! and Bumblebee from DC Super Hero Girls, who are both good. Here, she plays Panda Mania, a villain.
- Patton Oswalt, a comedian, plays Chameleon, a fairly serious-minded mercenary. His other villains are generally more comedic and over the top. When he voices good guys, they tend to be a Jerk with a Heart of Gold or Jerkass Woobie, not someone straight up nice like Uncle Ben.
- Promoted Fangirl: Felicia Day is a big fan of Mary Jane Watson, hence why she jumped at the chance to voice her here.
- Prop Recycling: The CGI backdrops appear to be reused from Ultimate Spider-Man, albeit very slightly modified with new locations (such as Horizon) and a different, more cel-shaded aesthetic.
- Recycled Script: The basic plot of "The Stark Expo" (entering an academic fair of some sort, and being pursued by a villain who wants the invention) is very similar to the Miles from Tomorrowland episode "The Unfair Getaway".
- Role Reprise:
- Robbie Daymond reprises the role of Spider-Man from the English dub of Marvel Disk Wars: The Avengers
- Travis Willingham reprises his role from the Spider-Man Unlimited game as the Sandman.
- "Halloween Moon" sees Fred Tatasciore once again voicing Hulk.
- In addition to the Hulk, Roger Craig Smith, Mick Wingert, Laura Bailey, Fred Tatasciore, Kathreen Khavari, Jim Cummings, and Mark Hamill reprise their respective roles from Marvel Universe shows as Captain America, Iron Man, Black Widow, Ms. Marvel, Crossbones, the Ghost, and Arnim Zola.
- Olivia Holt and Aubrey Joseph reprise their roles from Cloak & Dagger (2018) as the titular characters.
- The Brazilian cast features the voice actor Wirley Contaifer who voiced Peter in Homecoming and voices him as well on this series. Eddie Brock also also has the same voice actor (Philippe Maia) as the character's incarnation from Spider-Man 3.
- Schedule Slip: The first half of season 2 ended on August 13, 2018. The second half premieres in September 2019, which is 13 months later, or over 1 year.
- Short Run in Peru: "The Stark Expo" aired in places like South Africa about a week before the US.
- Show Accuracy/Toy Accuracy:
- The official Hobgoblin action figure from Hasbro looks nothing like the design featured in the show, instead sporting a green mask and purple cap similar to those of the classic Green Goblin costume.
- Most of the toys and promotional art depicting the Venomized Hulk have Bruce Banner be the one bonded with the symbiote, even though in the show it's actually Amadeus Cho.
- Spoiled by the Cast List: The villain responsible for the events of "Bring on the Bad Guys" is left unnamed, but the cast list revealed him to be played by Scott Menville, the voice of Doctor Octopus, making it easier to guess who it was.
- What Could Have Been:
- An early promotional poster◊ gave the show a more comic book look to it. Dr. Octopus and Rhino resembled their comic book counterparts more, and Vulture's costume would have been his Spider-Man: Homecoming suit.
- The ending to "Generations" was slightly changed, according to storyboard artist Kelly Kao. The episode shows only Peter meeting with Harry at the diner, unlike the storyboards which also show his friends coming in a bit later.
