Debut: Battle Scars #1 (January 2012)
Voiced by: John Eric Bentley
A super-spy and natural-born leader who sits at the head of the global defense organization "SHIELD", Nicholas Fury Jr. isn't afraid to jump into the fray and show those who threaten Earth why he commands the respect of Earth's greatest heroes.- Authority Equals Asskicking: The leader of SHIELD, but he's ready to jump into the fray if needed.
- Bald of Authority: Certainly counts considering he's inspired by his predecessor's Ultimate counterpart, who was in turn modeled after Samuel L. Jackson.
- Demoted to Extra: His father was a playable character in the first two games, albeit as a secret character that had to be unlocked. Here, there's no sign of him being playable.
- Eyepatch of Power: Missing an eye, but he is a veteran warrior and a great leader.
- Generation Xerox: Nick Fury Jr. is identical in almost every respect to his father aside from being African-American, and is also based on the Ultimate and MCU versions of Nick Fury.
- Only Sane Man: He's generally the one who steps in when things get out of hand between the heroes, such as keeping Iron Man and Rocket Raccoon from getting into a pissing match.

- Badbutt: Jessica doesn't get to be a Sir Swears-a-Lot here (though Rocket does allude to her "colorful vocabulary" in her profile), leading to awkward situations where she refers to the Hand Ninjas as "sneaky little jerks".
- Civvie Spandex: Jeans, t-shirt, leather jacket. She's a P.I., not a superhero.
- Comic-Book Fantasy Casting: A dead ringer for Krysten Ritter, her actress in the Netflix series. By extension, it causes an Adaptation Dye-Job.
- Happily Married: It's clear she and Luke love each other deeply.
- Punch-Clock Hero: To a degree. She actually bows out of the mission against the Hand because a better playing client calls her with an emergency.
- Private Detective: Her chosen career, as being a superhero didn't work out for her.
- Small Steps Hero: She states that she joined the Defenders because somebody needs to look out for the average citizens of New York when the A-list heroes like the Avengers are usually off fighting aliens on the moon.
- Super-Strength: Her primary superpower.
- Took a Shortcut: She manages to find a way ahead of the heroes. When questioned how she got there so fast, she says that it's a secret.

This sapphire scientist has friends with formidable facilities all over our world... and on countless others!
- Adaptational Modesty: Forgoes his famous trunks for shorts.
- Amazing Technicolor Population: A blue-furred man.
- Beast Man: Well, he's clearly meant to look like some sort of animal, but as his design takes from the 90s era, it's a bit difficult to tell what exactly he's supposed to evoke.
- The Bus Came Back: This marks his first appearance in the Action-RPG games— playable or otherwise— since the second X-Men Legends game.
- Composite Character: Seems to take after his 90s ambiguous creature portrayal, but wears a costume more in line with his more modern designs.

- Guest-Star Party Member: Serves a support role as a Mr. Exposition.
- Handy Feet: Like Nightcrawler his feet are like an ape's, allowing him to perform amazing acrobatic feats.
- Nice Guy: One of the politest members of the team, often serving as a voice of reason, logic, and morality.
- Sesquipedalian Loquaciousness: He's very well-spoken and prone to peppering his speech with unnecessarily large and profound-sounding words.
- Walking Shirtless Scene: Well, he IS covered in blue fur so there's no real point.
Blackagar Boltagon / Black Bolt
Species: Inhuman
Title: Inhuman king. The strong and silent type.
Debut: Fantastic Four #45 (December 1965)
Voiced by: Liam O'Brien
A member of The Inhumans' royal family from their floating home city of Attilan, Black Bolt was exposed to Terrigen Mist while still in the womb, thusly born with the incredible (and destructive) power of a hypersonic voice. He can also fly, manipulate particles, and has displayed limited telepathy. Because of the aforementioned hypersonic voice, Black Bolt was forced into seclusion for most of his upbringing, training in a soundproof chamber to control his power.
- The Bus Came Back: Was absent from the second game, but reappears in the third.
- Cain and Abel: The Abel to Maximus' Cain.
- Elective Mute: As usual. Justified— see Power Incontinence below.
- The Good King: Cares about his people more than anything in the world, and is always ready to defend them to the last stand.
- Super-Scream: Black Bolt's most devastating offensive weapon and one of the phenomena based upon the particle interaction is his actual voice. His voice is so powerful that even a whisper could cause catastrophic amounts of damage, and thus he refuses to speak and is so mentally disciplined that he doesn't even talk in his sleep.
- Power Incontinence: Incapable of speaking as even a whisper is devastating in its effect.
- Team Switzerland: Remains neutral in the conflict, only being concerned with the safety of Attilan and his people. Past events between the nation and Earth have also strained relations.

Species: Human mutate
Title: War hero turned weapon. Built to last.
Debut: Captain America Comics #1 (March 1941)
Voiced by: Ray Chase
James Buchanan Barnes, better known to his friends as "Bucky", was a young soldier who fought alongside Captain America as his sidekick until his apparent death in 1945. As fate would have it, he, too, was frozen in ice and subsequently thawed by a national superpower to become a super soldier — only in his case, he was revived by the Soviet Union. In time, however, the Winter Soldier would join forces once more with Captain America as one of the world's protectors.
- Adaptational Heroism: In the original game, the Winter Soldier was a boss working for the Masters of Evil; it's also implied that he was willingly working with them even after recovering his memories. Now he's an ally to the heroes.
- Artificial Limbs: Having lost his arm in the explosion that supposedly killed him, he was outfitted with a prosthetic arm after being picked up by the Soviets.
- Bash Brothers: He was this with Captain America back in World War II when he was still just Bucky, and even after becoming the Winter Soldier, the two still occasionally team up.
- Brainwashed and Crazy: His becoming the Winter Soldier is due to being brainwashed by the Soviets. As such, when in Wakanda he can't support the team directly as M.O.D.O.K. probed his mind to discover the code words needed to compromise him.
- Cool Mask: One that covers his entire lower face.
- Human Popsicle: He was kept in cryo-stasis when he wasn't on an assignment for the Soviets.
- Older Than He Looks: He looks like a man in his twenties, but he was actually a soldier in World War II.

- Badass Teacher: He's the headmaster for the Xavier Institute.
- Mission Control: Becomes this in the second DLC as well as the Danger Room challenges.
- Pet the Dog: He'll make plenty of compliments to teams during the Danger Room challenges like praising the Avengers for being worth the title of "Earth's Mightiest Heroes" or that the Marvel Knights are talented despite their anti-heroic nature. Amusingly, the one exception to this are the Inhumans (only heard if Crystal and Ms. Marvel are on the same team), he'll make backhanded compliments at best and mock them at worst.
- Psychic Link: He often speaks through telepathy instead of the comm like the other heroes.
