  • Ganke being able to piece together that Peter is the original Spider-Man in the game's introduction because of how he and Miles interacted, especially when you take into consideration that Peter was only his teacher for roughly two minutes.
    Spider-Man: Knew I never should've taught at a gifted school...
    • Made even funnier by how it takes Pete a solid three seconds for him to realize the significance of Ganke referring to him by his real name.
  • One of the missions in the aftermath of Sandman's attack has Miles rescuing an unconscious J. Jonah Jameson from a limo and taking him to the hospital. Jameson then proceeds to come to in the arms of said hero mid-Building Swing. He takes it about as well as you'd expect by launching into one of his trademark anti-Spider-Man rants without skipping a beat.
    Jameson: But J. Jonah Jameson is not going anywhere, you hear me?! I'M NOT GOING ANYWHERE! [notices the stretcher he's on is being pushed into the hospital] ...Wait, where am I going? WHERE AM I GOING?!
    • The fact that none of the Spider-Men or the paramedics give him a Shut Up, Hannibal! or even explain to him that they’re saving his life (Miles does correct him once) just goes to show that he is so delusional that he is not worth talking to anymore.
  • Spider-Man spends one of the tutorial fights with a group of thugs stealing guns from a gun club complaining about how he doesn't see the purpose of a gun club in a city like New York beyond something for criminals to steal from and begins suggesting more socially beneficial replacements for the club, all while continuing to beat up the criminals.
  • With the game's inclusion of fall damage it's possible to mistime a web-swing and cause Peter/Miles to pancake on the concrete below in a manner as wincingly painful as it is absolutely hilarious. Doubly so if you try to do a loop de loop and faceplant on the roof of a building behind you. You get an achievement making fun of you the first time you do so.
  • In the Midtown High flashback, teen Peter and Harry must slip past security guards. Harry's distraction so the cornered Peter can escape? Sing the chorus of "Never Gonna Give You Up" on the speaker system set up for Homecoming. Even Peter can't help but chuckle at the fact that Harry Rickrolls the cops!
    • Also in the flashback, in the photo op room, as Peter notices a picture, he recounts his first meeting with MJ, and apparently didn't know whether to say "hi" or "hey". So he decided to say "Hiyay".
      • If playing with Swedish subtitles, this becomes yet a little funnier. Said subtitles say that Peter was torn between "tja" ("hey") and "hej" ("hi") and wound up saying "tjej" ("girl") instead.
  • While Peter's fight with Scream is very heart wrenching, there's a bit of dark humor when at one point they tumble onto an ice cream truck that says "we all scream for ice cream".
    • Right after the fight, Jonah calls MJ, whom the young lady has had enough of and tells him THIS:
      Jonah: WATSOOOOOO- [gets cut off by MJ hanging up].
      Peter: You're so cool.
  • After Miles sends Felicia to Paris, he tries to use the magic portal wand stick to find Li, only for it to vanish and be replaced with a simple IOU apology note from Wong, much to the web-head's slight annoyance.
  • At Coney Island, you can end up getting a prize after getting enough tokens; some combination of ostentatious large glasses and hat, some with neon lighting, even. Shortly after the Hunters attack, Peter will change into costume and charge into action.... While still wearing the hat, which stays on as long as you don't die.
  • When the Hunters raid Coney Island to abduct Tombstone, MJ notes that they're not even remotely interested in going after Mysterio. You can just imagine Mysterio casually shrugging his shoulders and watching the show unfold.
  • After discovering Harry's Venom powers, Peter and Harry spend a night at the Emily May Foundation experimenting with them. Harry seems to have some issues controlling his abilities which leads him to break a few things. Cut to Norman walking in to find the office completely trashed, with both Harry and Peter fast asleep on the couch. One wonders what Norman's first assumption was as to how the room ended up in that state.
    • His almost deadpan delivery of “What the hell happened here?” makes it even funnier. It gives the impression of a parent coming downstairs to find the aftermath of their kid’s sleepover. Which is almost exactly what’s happening.
  • Harry's enthusiasm about being a superhero and his lack of experience lead to a lot of good moments when he joins Peter to rescue Tombstone from the hunters.
    Spider-Man: There's our guy. What's the plan?
    Harry: I think we should get him.
    Spider-Man: [Visibly baffled] Wh-
  • During the Hunters’ raid on the Emily May Foundation, Peter is confronted by a brute wearing a bearskin. Peter tells him, very seriously, that he has something to tell him before they fight. When he gives his message, you can just hear the shit-eating grin on his face under the mask.
    Spider-Man: [stage whisper] I’m fresh out of honey.
  • During Venom's rampage in the Oscorp building, he gets the all too familiar prompt of a wall grate, which Spider-Man usually uses to slip into the air vents and sneak around. Seeing as Venom is a giant compared to Peter and Miles, his attempt to zip up into the grate just has him smash through the wall.
  • The entire flashback to Peter's first meeting with Jameson, including:
    • Peter tries to say that with Uncle Ben's death and the mortgage being due, he needs the money, leading to Jameson stating that he does not care for Peter's sob story.
    • In typical Parker luck, his trip to the Bugle involves having to weave through traffic, a truck dropping several boxes and fish on the ground, construction up ahead, and culminating with him having to intervene in a robbery. Although that last moment also doubles as a Moment of Awesome, since that basically gives a playable combat segment as an unmasked, unsuited, younger Peter.
    • Peter's dialogue during the fight shows he's got a lot to learn about quipping.
      Young Peter: Hey...what are you, a tourist slack-mouth? Kidding!
      Young Peter: This is gonna hurt! But not, like, permanently!
      Young Peter: Hey ugly! Like, under your mask, probably!
      Young Peter: The bigger you are, the— Not that you're fat, I didn't mean that!
      Criminal: Now I gotta hit you harder for being so BAD at that!
    • While Peter complains about the traffic, Jameson advises him to shout "Look out, halfwit! These are my streets!" like the New Yorker he is. Cue the Inopportune Voice Cracking from Peter:
      Young Peter: Hey, halfwit— oops, sorry, 'scuse me! Scooting by!
      Jameson: Ugh. Parker, pathetic! Be a man about it! SHOW THEM YOUR MANHOOD!
      Young Peter: Yes, sir—wait, WHAT?!
    • Jameson casually remarking that Peter will get himself killed trying to make it to the Bugle.
    • And when Peter finally makes it to Jameson and shows off a picture of Spider-Man, Jameson remarks that his lighting is flat and his composition is awful, plus he's shooting on film like a caveman and his nose is bleeding from getting involved in a fistfight with a mugger... only to hire him because he needs photos of Spider-Man.
  • While planning for the final mission, the team uses pieces from a child's board game to put on a cartoon war map to do so. For example, they use a big goblin to represent Venom, a stereotypical hero for Peter, a bunch of baby goblins for all the symbiote spawns, and a rock that they stuck googly-eyes on as the meterorite. The goblin pieces being used for Venom and his symbiote-army gets even funnier considering how there is an Osborn in that black suit.
  • In front of the Oscorp building there's a group of protestors demanding that Oscorp and Norman Osborn face justice for their involvement in the events of the first game, specifically the bioweapon that was unleashed on New York... except for two people who have signs saying "Norman doesn't tip!"
  • One of the Photo Ops in Central Park is a conspiracy theorist who's going off about... aliens invading New York! The protagonists admit it wouldn't be a surprise, given the kind of world they live in.
  • After activating The Flame side quests, Peter and Miles can run into Wraith during crimes. If you interact with her after a battle, the Spider-Men will attempt to either fist bump or high five her... only for her to remain unresponsive and stare at them, which they respond to by sheepishly shrugging her off, as if to say, "Fine, whatever, be like that..."
    • The first time you encounter her as Miles, he awkwardly greets her as "ma'am", which Wraith is audibly indignant about. Seems that even violent vigilantes aren't immune to the Ma'am Shock.
  • Wraith's lack of response to any humor leads to a moment at the beginning of the Everything Burns mission when Peter cracks a quip and almost immediately regrets it.
    Wraith: They care a lot about... the "Crimson Hour"... Whatever that is.
    Spider-Man: The hot new club in Hell's Kitchen?
    (Beat)
    Spider-Man: [Voice noticeably shaky] Wow, that... just... popped out there, huh? Who says stuff like that? Not me.
  • Jameson ends up stuck in a traffic jam following a wrecked tunnel, forcing him to record his podcast in his car.
  • Peter’s fight with the symbiote possessed MJ takes on a darkly humourous turn when the whole fight looks like a marital spat. One considers that bystanders witnessing and listening probably were thinking; “I know couples have fights, but damn!”
    Scream: YOU can’t keep a job. YOU can’t pay the mortgage.
  • If Peter and Miles finish a fight together, they’ll do a random action afterwards, when prompted. Sometimes they’ll bro-hug, other times they’ll high five, and of course they’ll point dramatically at each other.
    • If you run into Harry during crimes while he's Agent Venom, Peter and Miles finger gun with him multiple times after the battle in a funny and endearingly enthusiastic way. Even funnier is that after this action, Harry never moves and stays in the same spot until you run into him again, as if he's thinking, "Okay...what do I do now?".
  • When Kraven's forces attack on a prisoner convoy carrying Scorpion and Mister Negative, during the ensuing fight some of the goons might shout to target Miles, "the little one" first, which offends Miles.
    "Little one?" You better be talking about the other Spider-Man, because I'm a half-inch taller than him at least!note 
  • Peter's exclusive dive trick is solving a Rubik's Cube in midair and just like in Miles Morales, he can definitely crash into the ground (or even die with Falling Damage). For bonus points, use the Spider-Man Noir suit.
  • During one of The Flame missions, Peter will ask Yuri if she has any tattoos. Her response?
    Wraith: Just the one of Spider-Cop's gravestone.
    Spider-Man: C'mon, Yuri. Really?
  • It's in the backdrop of an otherwise heartbreaking scene, but there's something darkly funny about Peter and MJ silently pulling a Screw This, I'm Out of Here! from the reception room while Norman is smashing things with Harry's cane in the final scene where Harry is confirmed to be in a coma.
  • Through the use of a glitch, you can free roam as Venom. This includes hilariously goofy animations for swimming and even riding a bike. Even funnier, there are actually a few NPCs walking around, and they don't react to him at all, seemingly oblivious to this 9-foot tall monstrous guy running around the city.
    • Another glitch allows Venom to become fully playable. Because Venom can grab enemies, including Brutes, which are not programmed to have an animation when grabbed is a sight to behold.
  • While the fight against Peter is extremely hectic and emotional, if you die during the battle, he'll sometimes do a melodramatic cheer with his hands as if he's saying, "Yay, I win!", which is so jarring that's it's difficult not to laugh at.
  • During Spider-Man's attack on Kraven's first compound, one of the the Hunters crows that "Kraven hunts what man fears most!". Peter is not impressed.
    Spider-Man: Oh, so he hunts failure? Intimacy? Clowns? If it's clowns, good riddance, I say!
  • Remember the drunk shirtless dude wearing a Spider-Man mask from the first game? You can encounter him again here, still shirtless and drunk and having a photo op done with a second shirtless, drunk guy, the latter wearing Miles' Spider-Man mask.
  • After drugging Kraven's pet tiger, Dima, so she won't try and eat him, Peter is given the option to pet the snoring tiger, and he even gushes and coos over her like she's a kitten and not a wild animal that tried to kill him. If you're curious what happened to Dima after Kraven's death, you'll find her in the Central Park Zoo, where she's been apparently seized by the state and placed in the tiger exhibit there. You can pet her there too.
  • During the post-game, you can encounter a reporter being filmed at city hall, and if you approach her, she'll give you a quick interview.
    Reporter: Spider-Man! New Yorkers want to know... is a hot dog a sandwich?
    Spider-Man: No, but it might be a salad.
  • Both Peter and Miles can join in a yoga session with some civilians.
  • There's a hidden event in which a random guy drops his phone in a manhole and asks for Spider-Man to get it out for him. Right after warning him to be more careful of his surroundings when using it, the guy proceeds to start texting in the exact same spot he dropped it near.
  • Since New Game Plus lets you use costumes within cutscenes where Peter usually has the black suit, if you wear something like Spider-Man Noir's outfit from Spider-Man: Spider-Verse, it can result in something like this.
