Hiroaki Uzuki, the Former Ultimate Apprentice. Who was he an apprentice to? Growing up poor, all he ever wanted was to be acknowledged by his workaholic father who never paid much attention to him. This along with the tarnished reputation of their family name spurred him to take on different jobs in an attempt to prove himself. Surprisingly enough, he proved adept at handling various tasks with incredible proficiency to the point of being branded an Ultimate. Hiroaki dreams of one day restoring the Uzuki family's honor using his newfound talents.

- Art Evolution: He and Bussho are the only characters to get revamped character designs to better match their respective talents.
- The Baby of the Bunch: He's the youngest child within the Uzuki family.

Ultimate Angler

Introducing Ikki Namiki, the Former Ultimate Angler. This young man can be best described as unpleasantly reserved, collecting his thoughts for a moderate period of time before speaking. Following in his father's footsteps, he became a phenomenal fisherman whose talents were soon noticed by Bitokukawa Academy. Not one to indulge in attention from others, he surprisingly gets chatty when it comes to shonen anime.
- The Baby of the Bunch: Is the youngest male student in the class.
- Berserk Button: Does not tolerate hypocrites.
- Big Ol' Eyebrows: Has the thickest eyebrows among the cast.
- Handicapped Badass: Despite having lost an arm, Ikki's fishing skills are far from diminished. He is also a skilled swimmer which makes sense considering the nature of his ultimate talent.
- Hidden Depths: Aside from being an accomplished angler, Ikki enjoys drawing and writing during his spare time.

Meet the Former Ultimate Pawnbroker, Kunitaro Higa, a vintage tech specialist with a keen eye for nostalgia. Having spent most of his years working at his grandfather's shop, he learned to appreciate hand-me-downs after years of being taunted for it during his childhood. Kunitaro is determined to establish himself as the top pawnbroker in Japan, and to make that happen, he willingly applies to Bitokukawa Academy as a student.
- Berserk Button: Hates it when people don't take responsibility for their own actions.
- Cool Shades: Meant to highlight his cool attitude.
- Informed Poverty: He grew to love things old school on account of growing up with hand-me-downs as a child and meeting customers who've had it rougher than him.
- Mellow Fellow: He's a cheerful man who tries to take things easy.
- New Technology Is Evil: Has a dislike for touchscreens.
- Outdated Outfit: Wears an outdated suit as his school uniform.
- Sharp-Dressed Man: While not as high-end as Oniji when it comes to fashion, Kunitaro's suit makes him fit this trope quite nicely.

Ultimate Yoga Guru

Bussho Saji is the Former Ultimate Yoga Guru. Sympathetic and soft-spoken, he has a tendency to act as the voice of reason for others whenever conflicts arise. Despite starting out as the pride of his family, Bussho was forced to fend for himself after his beloved mother died from poor health. As his life went downhill, he met a certain someone who inspired him to become the altruistic soul he now is.
- Ambiguously Brown: He has noticeably tan skin.
- Art Evolution: Bussho's character design was greatly modified to look less generic and more befitting of his status as a yoga guru.
- Berserk Button: Does not like anything related to politics. This makes sense due to the conflict-causing nature of politics as well as his own pacifist beliefs.
- Dark and Troubled Past: Went through a series of traumatic events that led him to leave his family behind upon graduating from high school.
- The Determinator: He does his best to ensure that his agendas are met.
- Disinherited Child: Implied to have become this after going through a tragic chain of events following his mother's death.
- Eyes Always Shut: Tends to close his eyes when in a good mood. Him opening them is usually a sign that something serious is about to happen.
- Momma's Boy: Really loved his mom when she was still alive.

Ultimate Forum Admin

Introducing Shoko Otaki, the Former Ultimate Forum Admin. To say Shoko has a short fuse is an understatement. With a zero tolerance policy against tomfoolery, he often expresses his loathing for it either through cursing or mockery. Because other kids would often tease him for his height, he found solace in the internet where he became extremely active in forums. Shoko's profound humility and patience as a forum admin earned him a place at Bitokukawa Academy.
- Air Quotes: One of his character busts has him do this.
- Berserk Button: Has a fit whenever someone makes nonsensical statements.
- Closet Geek: Loves action figures and card games.
- Expy: As a Tiny School Boy in serious need of anger management classes, Shoko is pretty much the Fuyuhiko of the game. He also shares Hiyoko's penchant for being aggressive due to their past as bullied victims.
- Gender-Blender Name: Shoko is a name usually given to young girls.
- Hair-Trigger Temper: His most notable trait or vice. Ironically, he's much more patient and understanding as a forum administrator.
- Hidden Depths: This current iteration of Shoko aspires to become a pianist someday despite being more skilled at forum administration.
- Older Than They Look: While most of the cast look much younger than they actually are, Shoko is the only one who resembles a grade schooler much to his resentment.
- The Napoleon: Is short in stature and shorter in patience. He has a complex about it due to being bullied by others in the past.
- Recurring Element: Regarding character design, he continues the DR theme of a character having light skin, black hair, and red eyes.
- Red Eyes, Take Warning: Has red eyes and is very aggressive towards anyone or anything that agitates him.
- School Bullying Is Harmless: Averted. Shoko being constantly bullied for his height has left with him some serious anger issues.

Yahto Stone Crown is the Former Ultimate Mayor, a towering man with incredible leadership and social skills. Having developed an interest in politics from a young age, he strives hard to gain the trust of many in hopes of bringing them joy. Although his accomplishments as mayor have earned him praise, some saw it fit to tear him down through slander and malicious rumors. Nonetheless, Yahto hopes that being a student at Bitokukawa Academy will help expand his talents like never before.
- Berserk Button: Hates journalists and bad manners.
- The Big Guy: Yahto fills the role as the designated "giant" student of the cast.
- Braids, Beads and Buckskins: Used to denote his Sioux background.
- Expy: Of Sakura Oogami. He has the Face of a Thug but is a genuinely good person and wants everyone to work together as friends.
- Foreign Exchange Student: One of the three foreign students of the class and the only male of that particular grouping.
- Friend to All Living Things: Along with his desire to form bonds with his survivors, he also likes dogs based on his report card.
- Genius Bruiser: Has a strong build and is mentally competent enough to run as mayor.
- Gentle Giant: Like past "Big Hearted Muscle" characters, Yahto strives to achieve trust among his fellow students to work together, but is fearful that he may not appear sincere because of his title.

Ultimate Tailor

Oniji Itagaki is the Former Ultimate Tailor. Smarmy and hotheaded with an overbearing personality to boot. Oniji makes it his business to look good and surround himself with people who complement his outward appearance. Born to a wealthy family of tailors, he harbors a deep resentment towards his father for putting constant pressure on him. As a result, Oniji became a narcissist with a materialistic streak to cope with the pain.
- Can't Take Criticism: One of his defining flaws. This may have something to do with his less-than-stellar family life.
- The Dandy: Obviously enough.
- Expy: His personality and appearance makes him similar to Kaito Momota only with a red/fire theme and an interest in fashion.
- The Fashionista: A rare gender-flippped example. Oniji has a fashion themed talent and believes that the right wardrobe is vital in making a true man.
- Fiery Redhead: A hothead with a snarky streak.
- Gratuitous Spanish: Sometimes peppers his sentences with Spanish words or phrases.
- Hates Their Parent: Hates his father with a passion.
- Hypocrite: Has no problem dishing out harsh criticism on others yet can't actually take them himself.
- Narcissist: Believes that good looks, good cars, and good company are what make a true man. He aims to embody these traits and won't settle for anything less.

Ultimate Stuntman

Manzo Shibue is the Former Ultimate Stuntman. Fun-loving and full of heart, he does whatever he can to make his loved ones happy. Ever since his father died from a car accident, Manzo developed an overprotective streak towards his mother and often looks after her. However, looking after himself became a rather questionable topic due to his growing interest in stunt performers. Though he and his best friend became stuntmen at the same time, only Manzo was chosen to be an Ultimate for Bitokukawa Academy.
- Adaptational Nice Guy: The renewed story portrays Manzo as more caring and less egotistical.
- Cool Board: Uses one in his splash art pose.
- Cute Little Fangs: One pokes out when he closes his mouth.
- Fragile Speedster: While fast and nimble on his feet, Manzo is often susceptible to injuries.
- I Want My Beloved to Be Happy: Genuinely wants what's best for his mother and friends, though this trait sometimes clashes with his occupation of choice.

Ultimate Theorist

The main female character of Kill/Cure. As befitting of the Former Ultimate Theorist, Hide specializes in making analytical deductions that are usually spot-on. Raised by her grandparents, she took up political science to follow in her father's footsteps as a career politician and later became engrossed in human philosophy. She received numerous accolades for her academic essays on the subject, but has yet to receive any response from her father so far. Often a harsh realist in most situations, Hide can be kind and empathetic to those who have earned her trust.
- Academic Alpha Bitch: While Hide's not much of a bitch, she did maintain high grades throughout her school life.
- Adaptational Nice Girl: This iteration of Hide is much less of a Jerkass thanks to the renewed story.
- Berserk Button: Liars seem to anger Hide most.
- Birds of a Feather: Despite being portrayed as rivals, she and Hiroaki both have fathers who've constantly neglected them.
- Can't Take Criticism: Or rather she can't accept having her own theories denied.
- Expy: She brings to mind Maki Harukawa due to her cold and blunt personality along with her dislike for socializing with her peers. Her voice is also fairly similar to Maki's portrayal in the English dub.
- The Fashionista: While people around her would never know it, Hide has a budding interest in fashion.
- Ice Queen: As the main female lead, she acts aloof towards the other students and has a tendency to verbally lash out whenever her arguments get challenged.

Maru Yokokume is the Former Ultimate Activist. She's spunky, eccentric, and knows how to make a scene. As an only child, Maru was spoiled rotten before learning the value of teamwork and cooperation through sports. After a tragic incident further changed her outlook in life, she decided to use her passion to stand up for the underdogs of society. Equipped with her trusty megaphone, this young lady takes every opportunity to get her message across.
- Girly Girl with a Tomboy Streak: Despite her feminine appearance, Maru is more into sports and social issues.
- Mythology Gag: Pulls off some of these during her interactions with everyone else.
- Her megaphone is an Ultra Despair Girls edition.
- The teaser shows that she has a phone cover modeled after Monomi.
- No Indoor Voice: Has the loudest voice thanks to her assertive personality and megaphone.

Ultimate Epidemiologist

Uta Kawagichi is the Former Ultimate Epidemiologist. Both preachy and dedicated, she strikes fear into every being, whether they be cellular pathogens or human souls. Being the daughter of a seafood restaurant owner, Uta is no stranger to hard work and dedication. One day, she withdrew from school due to her growing obsession with cleanliness. She soon found a way to hone it by joining a local church and adopting their beliefs as her own. In time, she became a prodigy in the field of public health which led Bitokukawa Academy to scout her.
- Absurd Phobia: Refuses to touch door knobs to the point of making any of the students open them for her.
- Birds of a Feather: Has this with Shoko because they became who they are due to other people bullying them for their unusual traits.
- The Fundamentalist: Uta likes going to church and is described as being difficult when it comes to changing her mind about certain things.

- Iconic Item: Is shown to carry two disinfectant spray bottles.
- Irony: Despite her profession as an epidemiologist which is often considered a scientific career, Uta is a strongly religious girl.

Momoe Soga is "The Soaring Lass"... errr... the Former Ultimate Local Hero! Momoe prides herself in performing "dangerous" heroics, such as saving cats from trees and... helping schoolchildren cross the street? Contrary to what most people assume, she doesn't agree with combative solutions. When role-playing as Soaring Lass, she can put on a brave face for a time, but easily falls out of character when one of her several phobias is triggered. She also feels responsible whenever her efforts fail to help someone in need.
- Absurd Phobia: Is constantly hindered by several different phobias.
- Actual Pacifist: Despite what her title implies, Momoe is against all forms of violence.

Stoic and rather lackadaisical at times, Naiomi Danjuma is the Former Ultimate Architect and one of the three foreign Ultimate students in her class. Raised by strict yet loving parents, she wanted nothing more than to make them happy. She decided to pursue a career in architecture to one day build a home her parents have dreamed about. Although Naiomi was initially hesitant to apply to Bitokukawa Academy, her family insisted that she take this once-in-a-lifetime opportunity.
- Black and Nerdy: Comes from Nigeria and has a thing for architecture designs.
- Character Tic: She plays around with pencils to relax herself.
- Hidden Depths: Likes to cook but hates cleaning up.
- Misery Builds Character: Although Naiomi was drilled into becoming a hard worker by her parents, it's made her rather wary of cleaning in general.

Ultimate Human Subject

Fan Fan Long is the Former Ultimate Human Subject and one of the foreign students in Nakagawa's class. A cheery and kind soul who has trouble saying no to others, the poor thing is often taken advantage for it. Fan Fan's body has developed strange physical effects due to various chemicals she's tested in the past; however, she doesn't particularly mind them at all and even seems happy to be of use to others. With a massive appetite and a cast iron stomach, Fan Fan will eat everything in sight without pause.

Although she nailed her official debut in "Waltzing in the Sunshine", her rise to fame ended after a botched performance left her partially deaf in both ears. Despite this setback, she hopes to get back to show business someday by improving herself as an Ultimate in Bitokukawa Academy.

Meet the Former Ultimate EXTREME Couponer, Chikuma Konishi! Unpredictable in nature and regularly inebriated on caffeine, this crazy couponer is an insuppressible ball of fun. Chikuma has an aggressive playfulness about her that makes her both hard to love and difficult to hate. Because of her unreliable parents, she had to take care of her two younger brothers while dealing with the family's financial hardships through coupon collecting. From jumping on her peers' backs for piggyback rides, to challenging them to boxing fights, she never seems to be out of energy.
- Acrofatic: Has no problem doing strenuous activities like jumping or boxing despite her large size.

A kind, doting teacher who patiently cares for the well-being of her students, although she can be blindly optimistic regarding them. Having graduated from the academy herself, Shiori has learned the values of virtue, believing Ultimate students to be the true future the world needs to change.
- Bookworm: How her talents as the Ultimate Instructor blossomed.
- Conveniently an Orphan: Shiori grew up as one, though that didn't stop her from finding success in the world of academics.
- Does Not Like Spam: She apparently hates meat.
- Expy: Shiori is similar to Chisa Yukizome being that she was a former Ultimate who now works as a teacher and cares deeply for her students.

Kokorosaru is an AI-controlled monkey that monitors the killing game. He identifies himself as the "Ultimate Counselor". At first impression, Kokorosaru is a kind and lovable monkey that claims he doesn't want anyone to play the killing game, providing motives to deter participants from doing so. However, whenever someone decides to kill anyway, due to some unfortunate event, he temporarily transforms into an aggressive and twisted form to host the class trials.
- Anti-Villain: Despite organizing the killing game, he claims not to want any murders and creates motives to discourage them.
- Berserk Button: Is said to have a twisted and dangerous form if any of the students end up dying. Counts as a Superpowered Evil Side.
- Expy: Of Monomi/Usami. Along with being main mascot characters who try to encourage the students to get along with each other, the teaser shows that he isn't very respected by the students.
- Maniac Monkeys: A zigzagged example. Even though he hates murder and pushes the students to not kill each other, he's still the self-proclaimed organizer of the killing game. And then there's what happens when a potential student goes ahead and kills someone.
