A boss taking up an entire stage. The point of the stage isn't actually defeating them, as they're invincible. Instead, you have to guide an extremely weak NPC to the end of the stage without you or them dying. Combined with stage hazards, the boss' ability to drain your health from across the room you're in, and other enemies, the stage is extremely difficult, but at least it has the liberty of being optional.
And you can't play the stage again if you lose, preventing you from getting that 100% completion award. Have fun!

A Flunky Boss, always with another specialized Awe (each of which has its own pattern of movement and attack) behind them. They're also capable of Interface Screw attacks. They also can go back to earlier points in the fight, resetting the field to what it was at that point. However, their actual health pool is quite small, not much larger than the higher health Mooks.
This would be you run of the mill boss battle with three opponents serving as the boss.
This boss could be a High-Altitude Battle with Shock and Awe.

An Eldritch Abomination and the Greater-Scope Villain of an Roguelike game. The players must, over the course of the game's main story, prevent the boss' minions and cultists from summoning it into the game's world via their rituals, as the boss will be almost unstoppable should it enter reality. The boss is encountered mainly through 'Avatars of Blood'; weakened versions of the Boss' true self, which can be defeated and banished to slow the countdown towards a permanent game over.
In combat, the boss' Avatars utilise blood-themed attacks and spells, such as sacrificing a fifth of its current health to cut your attributes in half, or forcibly draining blood from your character to heal itself. Additionally, it can fly or float about to avoid attacks - however, it must rest on the ground from time to time, and is vulnerable under these circumstances.
If the boss is summoned fully into reality due to the player's failure to defeat the boss' cult, a Hopeless Boss Fight results - the boss' true form is both all but immune to damage, never misses its attacks, and cannot be blocked. Upon the PC's death, a series of cutscenes play, in which the boss and its cult wreak worldwide chaos and slaughter any resistance left on the world, before forcibly converting any survivors into members of the cult. This game over finishes with the boss directly addressing the player, mocking them for their failure before permanently deleting the active save file, forcing the player to start again.
"I swear, Colonel, I did not shoot at you because you called my sister slow. I would not miss if I did."
