Marshal Moo Montana
The leader of the C.O.W.-Boys.

- Actor Allusion: He's not the first marshal Pat Fraley's voiced in a Saturday-Morning Cartoon (in fact he used the exact same voice for both).

- Strong and Skilled: He both a capable rider and one of the best shots in the series. Justified as he is The Leader and has to keep his skills up so he can lead the others properly during their missions.

Dakota Dude
The strongest member of the C.O.W.-Boys.
- Alliterative Name: Both parts of his name begin with the letter D.
- Amazon Chaser: He falls in love with the tomboyish Cowlamity Kate.
- Bare-Fisted Monk: He prefers to use his hands in a battle as opposed to a weapon.
- Berserk Button: Never threaten or disrespect Cowlamity Kate.
- The Big Guy: He is the strongest of the three bovine lawmen.
- Blue Is Heroic: He's blue and a brave hero as well.
- Brainwashed and Crazy: Dakota, after being defeated by Skull Duggery and his gang in "Skull Duggery Rides Again", ends up having his mind controlled by them.
- Charles Atlas Superpower: He can lift boulders and other heavy objects over his head with ease.
- Fatal Flaw: Wrath. He's usually quite calm, but on the rare occasion he loses his temper, something bad always happens. This is the main reason he was defeated and brainwashed by Skull Duggery and his gang in "Skull Duggery Rides Again".

- Freudian Trio: He's the superego to Moo's ego and Cowlarado's id.

Cowlarado Kid
The youngest of the C.O.W.-Boys.
- Alliterative Name: Both parts of his name start with the same sound.
- Always Second Best: Downplayed, as was very talented with his lasso but he was still unable to surpass Cowlamity Kate in this area.
- Berserk Button: Don't remind him that Dakota has a badge, but he still hasn't earned his yet.
- Can't Catch Up: He was both unable to reach Cowlamity Kate's lasso skills or Dakota's skills as a deputy.
- He's a literal case of this when he and his horse Jezebelle can't keep up with Cowlamity Kate's cousin Cowline and her horse Tornado in the annual horse race.
- Chivalrous Pervert: He's a bit of a lech, but still an honest lawman.
- Defeat Equals Friendship: Played with when Cowlamity Kate's cousin Cowline defeats both Cowlarado and his rival Swifty in the horse race causing them to put aside their differences.

Lillian "Lily" Bovine
The owner of Cowtown's saloon.
- Always Someone Better: She was always one step ahead of her rival Sadie Wowcow, who came to resent her over this.

- Damsel in Distress: She's captured by the Masked Bull in the game and has to be rescued.
- Damsel out of Distress: Lily at certain points when getting kidnapped will often put up a fight and even free herself.

- Everyone Loves Blondes: She cares very much for Moo.
- Fanservice with a Smile: She's a very pretty and cheerful waitress.

"Cowlamity" Kate Cudster
A tomboyish cowgirl and occasional ally to the C.O.W.-Boys. Very sweet on Dakota.
- Action Girl: She could hold her own in a fight pretty well.
- Always Someone Better: Her lasso skills completely eclipse Cowlarado's prodigious talent.
- Art Evolution: She starts wearing lipstick in the second season.
- Brainy Brunette: She's a pretty intelligent woman with brown hair.
- Cute Bruiser: Cowlamity Kate, she may be pretty but doesn't stop her from keeping up with the cowboys in a fight.

Buffalo Bull
The town blacksmith.

Cody
A young boy who hangs around with the C.O.W.-Boys.

- Badass Adorable: He has his moments of this.
- Brainy Brunette: He's smart enough to invent Moo Mesa's first telephone which is instrumental to helping the C.O.W. Boys defeat Mules Burne in the episode "The Wild Wild Pest".
- Child Prodigy: He was able to make a working telephone for others in Moo Mesa to use with ease.
- Happily Adopted: He has no known parents, but is shown to be perfectly fine living with Miss Lily.
- Hero-Worshipper: For Moo, to the point he wishes to become a deputy.

Tejua
A young Native American girl who happens to be J.R.'s niece.

- Girlish Pigtails: She has a pair of these.
- Girly Girl with a Tomboy Streak: She is both very feminine and very adventurous as well.

- Nice Girl: She's a genuinely nice person.
- Non-Standard Character Design: J.R. and Tejua were both drawn without ears, unlike the rest of the characters.
- Pink Means Feminine: She wears a pink dress.
- Plucky Girl: She's brave enough to lure out the "Hole In The Ground" Gang so they could be captured. Moo even compliments her on this afterwards.

Sheriff Terrorbull/The Masked Bull
The sheriff of Cowtown, who secretly has a double life as a masked crook.
- Big Bad: Of the Arcade Game.
- Big Bad Duumvirate: He and Mayor Bulloney are the main villains.
- The Brute: To Mayor Bulloney.
- Commuting on a Bus: He is exiled from Cowtown in the second season premiere, but returns for a few more episodes to continue conspiring with Mayor Bulloney.
- The Dragon: To Mayor Bulloney.
- Even Evil Has Standards: As he reminds Boothill in "A Sheepful of Dollars", even bad guys know you shouldn't clean up your gun while it's loaded or there are other people around.
- Evil Counterpart: To Marshal Moo Montana.
- Fiery Redhead: Sheriff Terrorbull counts because his hide is red and he's often very angry.
- Final Boss: He is the final boss of the arcade game, in his Masked Bull disguise.
- A Fool for a Client: Subverted in "Bulls of a Feather". When Sheriff Terrorbull is taken to court for his crimes as the Masked Bull, he doesn't have a lawyer but Judge Bulloney does the cross-examination and shows a good reason to deem the prosecution's witness unreliable.

- Karma Houdini: The closest he ever gets to a comeuppance is being forced to leave Cowtown.
- Karma Houdini Warranty: In the arcade game, however, he is thrown into Cowtown Jail at the end of the game.

- Meaningless Villain Victory: Sheriff Terrorbull attempts to cheat at the raffle in "Stolen on the River" to win the tickets to the Dixie Trixie, only to find that the boat has been hauled into dry land after the C.O.W.-Boys discovered it to be a front for Five Card Cud's thievery.
- The Only One Allowed to Defeat You: When he and his brother team up against Moo Montana, they argue over who kills their foe.
- Recurring Boss: Appears twice as a boss in the arcade game, in the first level and again as the Final Boss.

Mayor Bulloney
The corrupt mayor of Cowtown.
- Bald of Evil: He's shown to be bald whenever his hat is removed.
- Big Bad Duumvirate: He and Sheriff Terrorbull are the main villains.
- The Evil Genius: He's the mastermind behind the Masked Bull's schemes and the corruption in Cowtown.
- Fat Bastard: His weight is big as his corruption.
- Greater-Scope Villain: He's the main reason the Masked Bull is able to commit so many crimes in Moo Mesa.
- Karma Houdini: He never gets any real punishment for his conniving efforts in manipulating and pilfering from the denizens of Cowtown.
- The Man Behind the Man: Mayor Bulloney is the reason the Masked Bull commits his crimes and keeps coming back.
- Mayor Pain: He's a corrupt mayor.
- Purple Is Powerful: In a political sense. He's purple and is the mayor of Cowtown.
- The Unfought: He makes a brief appearance in the arcade game, but is never faced in a boss fight.
Boothill Buzzard and Saddlesore Scorpion
Two dim-witted outlaws who work for Bulloney and Terrorbull. They are a buzzard and a scorpion.
- Adaptational Badass: They are much more competent in the arcade game, which features both of them as level bosses.
- Alliterative Name: Both their names are alliterations.
- Bumbling Henchmen Duo: They are rather dim-witted and incompetent, and are never seen apart.
- Co-Dragons: They're both the main lackeys of Bulloney and Terrorbull.
- Death by Adaptation: Boothill Buzzard appears to die in the arcade game, as he falls over and has his spirit appear with a halo before flying away after he is beaten in his boss fight. The cartoon, in contrast, had him remain alive throughout the series' run.
- Evil Counterparts: To Dakota Dude and Cowlarado Kid.
- Feathered Fiend: Boothill Buzzard.
- Friend to Bugs: A literal case with Boothill Buzzard, whose best friend was Saddlesore Scorpion.
- Hates Baths: In "Boom Town or Bust", Boothill Buzzard gives not wanting to take a bath as his reason for wanting to leave before the flood comes.
- Lean and Mean: Boothill Buzzard.
- Never Learned to Read: They occasionally bring up that they never learned how to read or write.
- Scary Scorpions: Saddlesore is a giant villainous scorpion.
- Too Dumb to Fool: Boothill Buzzard is literally so dumb that he is actually immune to the hypnotic effects of Bovini's magic crystal.
- White Hair, Black Heart: Boothill Buzzard.
Jacques Le Beef (real name Barney Finkleberg)
A smooth-talking, handsome Frenchman who is really a con-man in disguise.
- The Barnum: He's a grifter who scams the people of Cowtown in order to make a profit.
- Bitch in Sheep's Clothing: He was only acting sweet toward the ladies so he could try and get away with stealing valuables.
- Chick Magnet: He's able to charm the women of Cowtown with ease.
- Everything Sounds Sexier in French: He was able to charm the ladies of Cowtown with his French accent.
- Fauxreigner: He's not really French. The accent disappears when Moo Montana confronts him and unmasks him.
- Light Is Not Good: He wears a white suit but is not an honest person.
- Sharp-Dressed Man: Jacque Le Beef wears a good-looking white suit.
Horrorbull
Sheriff Terrorbull's younger twin brother. Terrorbull broke him from prison as part of a plan to protect his secret.
- Identity Impersonator: He impersonates the Masked Bull to protect his brother's secret.
- The Only One Allowed to Defeat You: When he and his brother team up against Moo Montana, they argue over who kills their foe.
- Something Only They Would Say: He's the only one who calls Moo Montana "Marshal Meathead". It's how Moo realizes he's impersonating the Masked Bull.
- The Unfavorite: He says his mother liked his brother best.
Sadie Wowcow
A rival of Lily's who once tried to put her saloon out of business by starting one of her own.
- Always Second Best: She could never measure up to Lily's talents and resents her because of it.
- Beauty Is Bad: She's just as beautiful as she is deceitful.
- Driven by Envy: The main reason she teams up with Mayor Bulloney is to out stage Lily.
- Dude Magnet: She got the attention of every male in the Tumbleweed during her entrance. Even Moo was temporarily enamored when she approached him.
- Evil Counterpart: She's best described as a much more unscrupulous version of Lily.
- Evil Redhead: She's completely selfish and sadistic in her attempts to try to out stage Lily.
- Fiery Redhead: Much like Lily, she has her moments of this.
- Light Feminine and Dark Feminine: Lily Bovine and Sadie Wowcow respectively are a good-natured woman and a manipulative rogue.
- Manipulative Bitch: She's willing to do anything in order to achieve her goals.
- Ms. Fanservice: It makes sense considering she's Lily's Evil Counterpart after all.
- Proud Beauty: A literal case to contrast to Lily's Humble Heroine, as arrogance is one Sadie's defining characteristics.
- Right-Hand Hottie: She is very attractive and is in league with Mayor Bulloney.
- Rival Turned Evil: She aligns herself with Mayor Bulloney so she can have a chance to get back at Lily.
- Shadow Archetype: She represents what Lily would be like without any standards or scruples to achieve her goals.
- Significant Green-Eyed Redhead: She has red hair and green eyes which reflect her fiery personality and her jealousy of Lily Bovine.
- Wasted Beauty: She's beautiful, but also completely untrustworthy.
Skull Duggery
The skeletal ghost of a greedy miner named Tom Duggery who has fought the C.O.W.-Boys twice.
- Back from the Dead: Tom Duggery who is resurrected as Skull Duggery.
- Came Back Wrong: Downplayed, as Tom Duggery he wasn't exactly a decent person, but after his resurrection he becomes a complete nightmare.
- Dem Bones: He's a skeleton.
- Fatal Flaw: Greed: Tom Duggery. In fact, it was Tom Duggery's greed and selfishness of his silver mine that got him trapped inside and eventually killed during a cave in, which caused him to return to the living world as Skull Duggery.
- From Nobody to Nightmare: A literal example when the greedy miner Tom Duggery was revived from death, he became a vengeful ghost with spectral powers.
- Knight of Cerebus: He's one of the most dangerous villains in the series.
- Meaningful Name: His name is a play on the word "skullduggery" which means underhanded or unscrupulous.
- Posthumous Character: Tom Duggery, who died in a cave-in years before the events of the series and is resurrected as Skull Duggery.
- Rapid Aging: Skull Duggery and his men turn several denizens of Cowtown into senior citizens (including Buffalo Bull and Cowlamity Kate) in "Skull Duggery Rides Again".
- Red Eyes, Take Warning: He's a villain with red eyes.
- Time Master: Skull Duggery and his friends were able to make buildings become dilapidated and to make people rapidly age.
- We Will Meet Again: Both of his episodes have him swear revenge after being defeated.
- Would Hurt a Child: He had no problem trying to drown Cody, Carly, and Jacob when he caught them exploring his mine.
Bat Blastagun
A bat bandit.
- Alliterative Name: Both of his names start with the letter B.
- Bat Out of Hell: He's a giant bat and evil.
- Dark Is Evil: He's villain who wears a dark outfit.
- Named After Somebody Famous: Bat Blastagun's name is a play on Bat Masterson.
- No Celebrities Were Harmed: His voice is a Jack Nicholson impression, not unlike Neil Ross' other role as Shipwreck in G.I. Joe: A Real American Hero.
- Pungeon Master: He makes a lot of bat and fang related puns in his appearances.
- Red Eyes, Take Warning: He's a villain with red eyes.
Shock Holiday
A masked bandit who uses lightning guns to commit crimes across Moo Mesa.
- Curtains Match the Windows: When he's unmasked he's revealed to have brown eyes to match his brown hair.
- Dark Is Evil: He's a villain with a dark outfit.
- Evil Sounds Raspy: He has a very raspy voice.
- The Faceless: Subverted, as he's just wearing a mask to cover his entire face.
- Large and in Charge: He's considerably bigger than his gang is.
- Lightning Bruiser: Shock Holiday appropriately enough is a big man and also surprisingly quick.
- Named After Somebody Famous: Shock Holiday's name references Doc Holiday.
- Shock and Awe: He uses lightning guns to attack his enemies.
- Small Role, Big Impact: Shock Holiday only appears in the first episode of Season 2 but the bet over who catches him is the reason Sheriff Terrorbull is forced to leave Cowtown and stays out for the rest of the series.
- Unusual Eyebrows: When he's unmasked, he's revealed to have a pair of these.
Longhorn Silver
A bovine pirate who tried to rob a train in Swampy Cove.
- Affectionate Parody: Of Captain Hook from Peter Pan.
- Long John Shout-Out: His name's an obvious nod to Long John Silver from Treasure Island.
- Pirate Parrot: He has a parrot.
- Purple Is Powerful: He's has a purple hide and is a capable fighter.
Billy the Kidder
A goat crook who has a fondness for practical jokes.
- The Prankster: Of the villainous variety. He pulls pranks because he likes to cause harm.
- Punny Name: His name is a play on Wild West outlaw Billy the Kid and the fact that he's a goat who likes practical jokes.
Dr. Wolfenstein
A mad scientist wolf who lives alone. Bat Blastagun conspired with him in "How the West was Shrunk".
- Dr. Fakenstein: His name is a blatant play on Frankenstein.
- Mad Scientist: He's a mad scientist.
- No Celebrities Were Harmed: He sounds an awful lot like Boris Karloff.
The Great Bovini
A ringmaster who owned a crystal he used to hypnotize people.
- Evil Sounds Deep: At times he could reach this point.
- Mind-Control Eyes: He has a stone that he uses to hypnotize people into stealing for him. The stone shrinks the victims' eyes. Boot Hill Buzzard, whose eyes are already that small, isn't affected.
- Repulsive Ringmaster: He's an evil ringmaster.
Five Card Stud
A villain exclusive to the arcade game, and the boss of the Minecart level, where the heroes fight him in his saloon.- Canon Foreigner: Has not appeared in the show, and only in the game.
- Con Man: His cry upon being defeated is that he doesn't have any money, implying that he plays crooked card games and cheats.
- Death Dealer: He attacks the heroes with floating patterns of deadly playing cards.
Spider Boss
An unnamed anthropomorphic Spider boss that appears in the game, as the boss of the Sheeza Butte level.- Giant Spider: A giant anthropomorphic spider that actually dwarfs the heroes in size.
- No Name Given: He was not strangely not given a name, and is the only boss to not have a name subtitle on his splash screen.
