Josuke and Okuyasu are a bromance. ExplanationOkuyasu quickly becomes Josuke's close friend and sort of brother-figure, but with some fans interpreting their relationship as a more intimate one, jokes abounded of the "bromance" culture.
And so began a beautiful friendship. ExplanationKoichi first meets Okuyasu after Okuyasu, initially an antagonist, slams Koichi's head into a gate, and later Josuke defeats Okuyasu by strangling him until he passes out. The fact that they become the closest and truest of friends later on despite this violent introduction was widely parodied by the fandom.
Josuke's fear of turtles is quite memetic in the fandom. JoJo's Bizarre Adventure: Golden Wind made this Hilarious in Hindsight after Polnareff's soul becomes trapped in a turtle's body after his death, leading to jokes about Josuke being terrified of Polnareff.
DuwangNoteAn infamous "Blind Idiot" Translation of part 4, which was originally done by a Chinese student for his English class project. The name Duwang comes from the translation of the name of the town Morioh, since in Japanese the name is written as 杜王町 and is pronounced "Morioh-cho". These same characters can be read as "Duwang ting" in Chinese. The improved Invincible Trio translations in the making keep a few of the more memorable lines as in-jokes and the TV anime is even being "translated" in this style by one fansub group.

Even crazier is how, despite sounding like a product of the notoriously shoddy translation quality of Duwang, it's quite accurate to the original Japanese. The original panel even has the kanji for "erection" and crude speech patterns inside of that very same speech bubble.
"It gave me a ROCK HARD COCK/ROCK HARD [REDACTED]"ExplanationThe official dub line for the scene. It was censored on the Toonami broadcast, likely due to the sexual nature of the line, but many found the use of the bleep funny. DC Douglas later confirmed on Twitter that the word that was censored was "cock".

Made even more memetic in that, out of all possible choices, the pose he makes with Crazy Diamond when he summons it to punch the spaghetti, is for some reason the go-to pose for him in most JoJo media◊.
Kinkshaming KiraExplanationYoshikage Kira is a serial killer who kills women and keeps their hands as "girlfriends" due to essentially having a fetish for hands. Thus the joke that the whole point of the protagonists defeating him in Part 4 is to shame him for his sexual interests.
"Dr. Jotaro Kujo: Respected 28 year-old marine biologist with a PhD., married man, and father to a six year old girl. Goes to Japan to sort out his grandfather's inheritance and almost immediatelygets into a fist-fight with a teenager." ExplanationHe accidentally insulted Josuke's hair and threw a pre-emptive punch when he saw Josuke summon Crazy Diamond, but to the non-Stand users present (i.e. Koichi and some of Josuke's female classmates), it looks like he punched a teenager for no reason. It doesn't help that he told the girls to shut up and stop crying, because it makes him come off as a Jerkass.
Jotaro vs. A Rat ExplanationOne episode has Jotaro and Josuke trying to hunt down a Stand-using rat, which has gained the ability to melt people's flesh into jelly. Jotaro is severely wounded by the rat, prompting jokes from the fandom that a little rodent actually came closer to killing Jotaro than even DIO ever did. A variation also mentions Stray Cat nearly killing Kira Yoshikage, joking that "Jojo animals are hardcore".
Koichi is the Polnareff of Part 4ExplanationHmmm, silver-haired sidekick who always suffers ridiculous indignities and is often the first victim of an enemy Stand? The two have quite a lot in common.
The joke gained traction when Koichi cuts his hair to resemble Polnareff's. Jokes have compared his small stature to the time when Alessi turned Polnareff into a little kid. Some fans have even started calling him Smolnareff!
With the reveal that Josuke patterned his hair out of gratitude to the mysterious teen who saved his life, fans have joked about Polnareff saving Koichi who then does the same.
The following part is book-ended by Giorno meeting these two (Koichi at the beginning arcs, Polnareff in the final arc).

Rohan's response of "But I refusenote Common fan translation/Like hell I will!note ASB translation/Daga kotowarunote Romanji/だが断る/Sorry, but I refuse!note Viz dub" Explanation This meme references when Highway Star commands him to call Josuke for help and thus lead him into a trap, is also a popular meme in the Japanese community, applied to any less than stellar Jojo-related info, product, or news, or even anything in general. Ironically, Rohan uses this line and claims that he likes to say "NO!" to the faces of people who think they're hot shit, despite Rohan himself fitting that bill a good deal. It's also seen as an act of badassery on Rohan's part, as he was in mortal peril and an inch away from losing his life to Highway Star only to rudely deny Highway Star's demand anyhow.

Explanation It's common for fans to insult Josuke's hair for laughs, which is almost always replied to by this very phrase, and thanks to All-Star Battle, a certain .gif◊ of this very phrase began to circulate as an appropriate reaction image.
The rain sounds like Josuke. ExplanationAfter Jotaro mistakes Josuke's screams for the rain, a small scale meme has cropped up around it. Such examples include reacting to Jotaro apparently having difficulty hearing, or depicting scenes of rain with Josuke screaming as background audio.
This is a case of Artistic License gone wrong. The manga has loud rain sound effects dominating pages to signify how loud the rain is, but the rain is softened in the anime so that dialogue can be heard clearly.
Memes have also been made about Jotaro damaging his hearing when he fired a gun next to his ear while in prison.
"The real protagonist of Part 4: ambulance." Explanation (spoilers)A joke making light of Kira's rather mundane death: accidentally backed over by an ambulance.
Green Super SaiyanExplanationIn his battle with Tamami, Koichi's hair stands up in spikes and a glowing green aura surrounds him, drawing comparisons to the Super Saiyans of Dragon Ball Z, particularly Gohan due to Koichi's small stature. Echoes Act 1 resembling Imperfect Cell has also not been lost on fans.

Giorno never met JosukeExplanationThe fact that Giorno never actually meets Josuke is commonly discussed in the fandom: the two have completely separate arcs, only sharing the fact that they both meet Koichi. As such, it's been frequently speculated by fans what kind of relationship the two would have, mostly them bonding over their hair vanity and desire to fight for good, though divided by Josuke's friendliness vs. Giorno's murderous ruthlessness.
The sharp contrast in tone between parts 4 and 5 is also widely memed, especially how Josuke tends to befriend and reform most of the minor antagonists of his part, while Giorno tends to mercilessly and brutally murder them, despite both of them on the side of justice.

Plenty of things involving Dub!Rohan:Explanation When Diamond is Unbreakable's anime got localized, notable voice actor Vic Mignogna was cast in the role of the mangaka and proceeded to give him a fittingly creepy and sleazy voice. At the time the dub was still going, Vic got hit with several sexual harassment allegations of which people are still torn on whether or not they're true. Despite the allegations having a huge negative impact on his career, the localization team kept Vic as Rohan throughout the dub's run, which only served to add even more memes to Rohan's resume.
Rohan’s manga has been pulled due to accusations of him hating kids.Explanation A joke playing off the controversial #KickVic movement when Mignogna was revealed to still be playing Rohan during the controversy, as the Janken episode aired in its dub form around the same time. Variations such as #GoBanRohan and #IStandWithRohan also began to spread like wildfire.

"I'm gonna fix that idol up!" Explanation The meme gained a resurgence in 2023 when Oshi no Ko started airing, with Josuke saving Ai's life.
Josuke will travel through the multiverse and save you from depression. ExplanationThanks to the edits of Josuke saving Saki from Metamorphosis, and followed by Ai, Josuke gains a reputation of a multiverse traveler, going through the multiverse of anime/manga, spot a depressing moment that involves a beloved sympathetic character dying and undo it by healing that character with Crazy Diamond, thus saving the depressed reader from those depressing endings. (Though in case of Ai, many other characters race to save Ai in their own ways)
"My name is Yoshikage Kira. I'm 33 years old." ExplanationWhen Kira's secret is discovered by Shigechi, he proceeds to launch into an extremely long and ridiculously elaborate speech which essentially amounts to an account of his daily life, all the while somehow managing to maintain an air of menace that prevents Shigechi from simply running away as he monologues. Because of the absurdity of the situation, this speech has been made into a copypasta.

Explanation Spoken by Kira as Shigechi discovers his little secret, and partially eclipsed by his more famous monologue right above, it has been used as a foreimage of He Knows Too Much on meme pages, like when someone sees the anime watchlist not having the first 2 parts, or when someone else discovers the browsing history.

Explanation Comparisons between Kars and Akira Otoshi abound, due to both having long purple hair, both having attempted to kill Joseph, and both being electric guitar players (Kars having mocked Lisa Lisa by pretending to strum her legs like a guitar in one famous scene in Battle Tendency.)
And then Josuke died of a broken spine.ExplanationThe very last time we see Josuke in the anime at the end of the final episode has Josuke striking a pose where he bends his body backwards. As he disappears entirely from the plot afterwards, not even being mentioned in Parts 5 and 6 and the resulting universe reset after part 6, some fans joked that Josuke just straight-up diedfrom posing and was never heard of again.
Kira triggering Jotaro Explanation Fans have noted several similarities between Jotaro's first personal encounter with Kira and the final battle against DIO in Part 3. In both fights, a wounded Jotaro is talked down by a blonde-haired villain who had just punched a hole through his green-clad friend's torso, with Kira even saying DIO's catchphrase. This led to jokes about Jotaro being afflicted with post-traumatic stress disorder from his past encounter with DIO, and Kira's eventual beatdown being the result of him unknowingly pushing Jotaro's Trauma Button.
Sukisukisukisukisukisukisukisukisukisukisuki! ExplanationWhen Josuke's mother Tomoko meets Jotaro for the first time, she mistakes him for Joseph and is so happy that she exclaims, "Suki, suki, suki, suki, suki, suki!" This has not gone unnoticed by fans, who joke about how the Stands should have had their names swapped.

Josuke burned down Rohan's houseExplanationDuring the 'I Am an Alien' arc, Rohan was so determined to figure out how Josuke cheated that he left a magnifiying glass near the sun, setting part of his house on fire. On the Shitpost Crusaders subreddit, it became a meme to portray Josuke as completely burning down Rohan's house or that he's an arsonist.
