- Eric Bauza as Puss, Eames, Senor Igualdemontijo, Desmondo (the donkey eared thief) & Wimblegurp. There are often scenes where Puss converses with Eames or Desmondo.
- Grey Griffin as Sphinx, Senora Igualdemontijo, Vina, and Duchess occasionally (see The Other Darrin below).
- Candi Milo as Kid Pickles & Cleevil.
- The Other Darrin: Eric Bauza takes over the role of Puss in Boots from Antonio Banderas. In a broader sense, Bauza also replaces André Sogliuzzo, who was Banderas' go-to replacement voice in most prior Shrek spinoff material.
- Duchess was voiced alternatively by either Maria Bamford or Grey Delisle.
- Uli was usually voiced by Alan Tudyk but Jim Meskimen voiced him for a few episodes.
- In his first appearance in "Sword," Goodsword was voiced by John Rhys-Davies. Following his return in "Sword's Man," Jeff Bennett voiced him for his remaining appearances.
- Production Posse: A lot of the people who worked on the show worked on El Tigre and The Book of Life, including voice director Andrea Romano, voice actors Eric Bauza, Carlos Alazraqui, Grey DeLisle Griffin, and Danny Trejo, as well as story editor Doug Langdale serving as executive producer and producer Jorge R. Gutierrez occasionally chipping in as a consultant.
- Role Reprise: Alexandre Moreno, the official voice actor of Puss in Boots in all of the films and specials in the Brazilian Portuguese dub, returns as the titular feline in the series.
- Voices in One Room: With a few exceptions here and there
(mostly when dealing with busy A list actors or large ensemble casts), the cast often records together, as per usual for most shows with Andrea Romano as the voice director.
