Possibly. They'd have to contend with the Assist Trophy version still being there since I don't think they'll just remove it (after all, Sakurai didn't bother removing Chrom from Robin's Final Smash), but there's ways around it like giving the playable version a different visual appearance (i.e. Assist Trophy Waluigi has the purple overalls, while playable Waluigi has his tennis uniform from Mario Tennis Aces).

Another Assist Trophy that I think could easily be Promoted to Playable: Akira Yuki.
His Assist Trophy uses his old, blocky, Virtua Fighter 1 model. If they make him playable, they could just use his more smooth appearances from later games.

To be honest, I’ll say no assist trophies will be made playable either when Sakurai explicitly says so, or the DLC comes to an end without it happening. I don’t think there’s reason to assume there’s an arbitrary rule saying it won’t occur.
You know, it's funny how Sakurai made a big deal about Grant Kirkhope being the first Western composer he's worked with, considering he then revealed not 20 minutes later that Toby Fox was the second Western composer he's worked with.
I won’t say it’s impossible, but they seem less likely. Maybe it’s my imagination, but Sakurai seems to think that being an AT is “close enough” in terms of a character’s representation.

It's also not some special honor any more. The first guest character was a huge deal but now than we're onto like the 15th who cares.
If the 5th fighter is 3rd party, then that will be the 15th guest character.

He and Dixie Kong are my two most-wanted characters now. As of September 4th, every single character I've ever wanted in Smash has made it, except for those two.

But anyway, Ganon (or Tetra) don't have to be the last Zelda character we ever get. It's not like there's a cap on how many characters each series can have. I just think Ganon would be a great new fighter, and we don't have any pre-Ocarina representation yet.
