Medusas
Chika Amagi
- Beware the Nice Ones: Normally she's a sweet girl who enjoys eating and cares about her friends, but once her murderous personality awakes, not many can escape from her unharmed.
- Big Brother Worship: Despite the favoritism from their parents, Chika was always close to her brother. And even Chika's murderous personality can't bring herself to kill him.
- Big Eater: Chika has a huge appetite. That she ate all the food Kiriko gave her as a form of hazing and still ask for more impressed the other inmates.

Sachiko Onigahara
- Important Haircut: She cuts her waist-long hair to shoulder length in order to allow Chika to briefly impersonate her during the SS Sanctuary arc and keeps the style afterwards.
- Light Feminine and Dark Feminine: She dresses in white, which contrasts with Chika's black uniform.
- Lipstick Lesbian: She's pretty feminine and attracted to girls.
- Master Poisoner: She's very good at creating concoctions to poison or incapacitate her targets.
- Putting on the Reich: Since her murder model was a Nazi, her Medusa costume is based on the white uniform of a nazi officer, Commissar Cap and all.
- Royal Rapier: Her weapon when not using poison is a saber, and while not a master swordsman like Aya, she's quite capable with it.
- Weak, but Skilled: She might be a deadly Master Poisoner, but she's not particularly strong for a medusa, leaving her at a disadvantage when facing physically powerful opponents.
Miina Isurugi
- Deliberately Cute Child: She uses her youthful appearance to pretend to be helpless in order to get her targets to lower their guards.
- Girlish Pigtails: Her default style and she's the most apparently innocent-looking of the Medusas.
- Monster Clown: Her costume is even named Mad Pierrot and it references that her murder model used to dress as a clown.
- Pædo Hunt: She has used her youthful appearance more than once to take down pedophiles who were trying to rape her.
- Put the "Laughter" in "Slaughter": She gets quite excited when killing, and she even uses blood to paint her face like a clown on occasion.
- Weak, but Skilled: Like Sachiko, Miina isn't particularly strong, so she uses tricks, traps and strangulation to beat her opponents.
Kiriko Murasaki
- Beautiful Singing Voice: As expected from a vocalist, it's been noted she has a beautiful voice.
- Curtains Match the Windows: She has reddish hair and eyes of the same color.
- Boxing Battler: Kiriko might prefer to fight with her Razor Floss, but thanks to her model she's also a deadly boxer.
- Hidden Depths: She has a license to cook pufferfish, which are deadly poisonous unless cooked by a professional.
- Razor Floss: Her primary weapon are strings which she keeps on her guitar and who she can use to strangle someone.
- Unexpected Virgin: Despite her previous claims, she was actually a virgin during the SS Sanctuary arc.
Karen Kirishima
- Alliterative Name: Karen Kirishima.
- The Big Guy: She's the tallest and the heaviest hitter among the Medusas.
- Combat Sadomasochist: The woman loves feeling and then inflicting pain, and it actually triggers her Charles Atlas Superpower whenever she receives a strong enough hit.
- Dye or Die: For the Shinsei Order Arc she dyes her blonde hair black to infiltrate the school.
- Jizzed in My Pants: Getting almost slashed from her womb to her belly makes her cum in ecstasy.
- Stripperiffic: Her outfit is just a bunch of straps that doesn't do a good job at covering her privates.

- Intergenerational Friendship: She forms a bond with Reika, a middle-aged janitor woman who's old enough to be her mother.
- Only Sane by Comparison: She occasionally plays the straight man to the other Medusas' wackier moments, but she's still a sex maniac and a serial killer.
- Promiscuity After Rape: She openly admits to Aya that she's a slut but getting raped by her father seriously messed her up.
- Psycho Knife Nut: Her weapon of choice are knives, which she keeps either on her thighs or hidden in her sleeves.
- Sympathy for the Devil: She admits she does have sympathy for Yuuri Nezu/Jeffrey Dahmer and her being unable to be loved right as she's burning to death while Katya watches.

Youko Kuroki
- Mysterious Past: Of all the Medusas, she's easily the most mysterious and whose past is explained at the latest.
- Psycho Knife Nut: Unlike Katya's throwing knives, Youko prefers to use a military knife in close quarters.
- The Quiet One: Alongside Aya she's easily the least talkative of the Medusas of Haguro.
- Scarf of Asskicking: Her Medusa outfit comes with a scarf.
- Tyke Bomb: She was raised since childhood to be a killer and an assassin.
Aya Tomoe
- A-Cup Angst: She has a modest chest size and is annoyed that Miina points out her breasts are larger.
- Hartman Hips: What she lacks in bust she compensates with wide hips and strong legs.
- Master Swordsman: Her weapon of choice is a katana and she's so fast and deadly she can cut someone to pieces within seconds.
- No Shirt, Long Jacket: Her Medusa costume has her wear a long jacket with no shirt or bra, occasionally showing her breasts.
- Obfuscating Disability: During the Shinsei Order Arc she pretends to be wheelchair bound in order to not arouse suspicion, but Madoka is capable of noticing she's faking.
- The Stoic: She's usually the calmest and most serious of the Medusas. When angry she usually goes through Tranquil Fury instead.
Seri and Maki Doujima
- Beware the Quiet Ones: Despite being usually quiet and insecure, Maki can be just as ruthless as her sister while in Medusa mode.
- Ninja Maid: Not really ninjas but the sisters dress in maid outfits as Medusas.
- Personality Swap: While in Medusa mode, Seri and Maki exchange their respective personalities unless one of them loses consciousness.
- Psycho Pink: Seri has pink hair and is just as dangerous and psychotic as the other killers.
- Shotguns Are Just Better: Just like their murder models, the sisters have always preferred shotguns to other weapons.
- Siblings in Crime: Much like their murder models, they're sisters who fight and kill together.
- Sibling Yin-Yang: Maki is quiet, meek and lacks self-confidence, while Seri is assertive, strong willed and outspoken. Even their color schemes reflect the difference, with Seri having bright pink hair and eyes while Maki has dull brown hair and eyes.
Haguro Prison
Kasumi Makube
The head female doctor of Haguro Prison Island.
- Everyone Calls Him "Barkeep": She's just known as Female Doctor for several chapters until her name is revealed.
- Hospital Hottie: Even Chika can't help but be captivated by her beauty.
- Morally Ambiguous Doctorate: Using girls as guinea pigs to turn them into serial killers and super soldiers is very much morally ambiguous.
The Warden
- All Men Are Perverts: The guy is a complete creep who masturbates at the sight of the girls.
- Fat Bastard: Overweight and a total bastard.
- Friends with Benefits: Appears to have this sort of relationship with the female doctor.
- Wardens Are Evil: Besides imprisoning girls to be used as guinea pigs for an nebulous MegaCorp, he's also a total pervert.
Nishi
- Amnesia Missed a Spot: Despite having his memory erased at least three times due to finding out the truth about the Medusas, the latest one fails to truly erase the memories, but chooses to keep quiet for his own safety.
- By-the-Book Cop: He's pretty straight laced and believes prisons should be used to rehabilitate criminals.
- Token Good Teammate: He's the only known staff of Haguro prison who actually just wants to do his job.
Azuma
- Flawed Prototype: She's a prototype Medusa, but how did she end up like that or what her murder model is is unknown.
- I Love the Dead: She's a necrophiliac and even keeps the pictures of dead women on her phone.
- Youthful Freckles: She has noticeable freckles and she's the youngest prison guard.
Civilians
Michitaka Amagi
The older brother of Chika Amagi, who uncovers the existence of Medusas and chooses to investigate the reason for their creation.
- Action Survivor: He's not a fighter, but he manages to escape with only minor wounds when he encounters the Medusas on Haguro island.
- Hidden in Plain Sight: He avoids detection by Itsubishi by simply taking off his glasses, changing his hairstyle and using aliases.
- Mr. Exposition: He's quite knowledgeable about serial killers and other subjects.
- Smart People Wear Glasses: He wears glasses and was actually studying in the US before he found out his sister had been imprisoned.
Akiho Kurumizawa
A journalist at Bunkyou Publishing Company who aids Michitaka in investigating the truth about the Medusa Syndrome and Itsubishi.
- Bespectacled Cutie: Wears glasses and is acknowledged as both cute and beautiful when serious.
- Drowning My Sorrows: Michitaka first meets her while drunk at a maid café, upset that her boss ignored her research concerning the Medusas.
- Ignored Expert: Her research into serial killing women was summarily ignored by her boss.
- Inherently Attractive Profession: She's a journalist and writer who happens to have huge breasts and attractive features.
Tendougumi
Tomomi Mizuno
The head of the Tendougumi Yakuza group. He really likes asses.
- Abusive Parents: He constantly anally raped his own son and is genuinely surprised she would betray him.
- Bald Head of Toughness: Completely bald and incredibly tough, capable of surviving a four story high fall with only a minor inconvenience.
- Bald of Evil: He's completely bald and a ruthless and depraved yakuza.
- Depraved Bisexual: Openly admits he doesn't care about the gender of his victims, if he likes their asses he will rape them.
- Even Evil Has Loved Ones: He seems to genuinely consider his lieutenants his family and mourns their deaths.
- Final Battle-Induced Shirt Loss: Loses his shirt during the final confrontation with the Medusas.
- Karmic Death: A man obsessed with asses who would anally rape his subordinates and who was convinced his ass would never fail him, dies by being impaled through the anus.
- You Have Outlived Your Usefulness: For trying to blackmail Itsubishi, stealing sensible information and since they no longer needed him, the Haguro Medusas were sent to kill him and any of his men who got in their way.
Niina Rudou
The son of Tomomi Mizuno, who dresses as a girl and is a big fan of Kiriko Murasaki.
- Elegant Gothic Lolita: Dresses in this manner.
- Girlish Pigtails: Wears their long hair in this manner, similar to Miina's.
- Nom de Mom: All There in the Manual mentions Rudou is their mother's last name.
- Sex–Face Turn: An unintentional example. Realizing he just took Kiriko's virginity makes him stop trying to kill her and immediately tells her he wants to defect from Tendougumi. Then they proceed to have even more sex.
- Unsettling Gender-Reveal: Kiriko was genuinely shocked to see the "girl" who was molesting her suddenly had a penis.
- Wholesome Crossdresser: Dresses as a girl and he easily turns on the Tendougumi due to his love of Kiriko.
Kyoko Kanzaki
A former pro-wrestler who works for the Tendougumi as an enforcer.

- Brawn Hilda: She's mannish, muscular and has two prominent scars on her face.
- The Dragon: She becomes Niina's main enforcer after she takes over ther Tendougumi.
- Even Evil Has Standards: Even she finds Mizuno's obsession with asses to be distasteful.
- Fiery Redhead: Color pages show she has red hair and is pretty eager and fierce in combat.
- Good Scars, Evil Scars: She has two rugged-looking scars on her face and she's not a pleasant person.
- Super-Toughness: The woman is incredibly difficult to injure or stop. Being shot at, hit in the head with a pipe or being electrocuted are only inconveniences for her.
Yan Bongfa
A Chinese woman and Kempou practitioner.
- Anime Chinese Girl: She wears a qipao and practices Kempou, a famous martial art, but her hairstyle and general personality don't fit the trope.
- Teeth Flying: Kiriko almost kills her by punching her in the face, breaking several of her teeth. She eventually recovers, sharpens them and turns them into Scary Teeth.
Shinsei Order
Madoka Sakuragawa
A Dakini, a Medusa created by the Shinsei Order. Her murder model is Edmund Kemper, the 'Co-Ed Killer'.
- Beware the Quiet Ones: She might be most of the time quiet and demure, but when in Medusa mode she's an utter beast.
- Huge Schoolgirl: She's pretty tall for someone the same age as Chika, which is fitting since her murder model stood at 6'4" in real life.
- Idiot Hair: She has a prominent cowlick and outside of combat she's rather absent-minded.
- Lightning Bruiser: She hits hard and she hits just as fast, and Aya notices she's not dumb either.
- Weapon Specialization: Her weapon of choice is a mallet, and she's strong enough to shatter walls and even decapitate people with her strength.
Yuuri Nezu
Another Dakini at the service of the Shinsei Order. Her murder model is Jeffrey Dahmer, the Cannibal of Milwaukee.
- The Collector: She collects the skulls of her victims along with pictures of them right before she killed and ate them.
- I Just Want to Be Loved: Like the real Jeffrey Dahmer, she desires for someone to love and understand her for what she is and that leads to her downfall, since it allows Katya to inject her with poison using a plastic caterpillar stuffed in her rectum.
- I'm a Humanitarian: Considering her murder model is one of the most notorious cannibalistic serial killers, it's no surprise she's one too.
- Four Eyes, Zero Soul: Wears glasses and even when not in serial killer mode she's heartless and unpleasant.
- Karmic Death: After lowering her guard around Katya, she poisons her and when she tries to retaliate against, she ends up tripping with the skull of one of her previous victims and ends up getting herself set on fire, burning to death.
