If you don't mind furries, TwoKinds includes a romantic relationship of that kind.
I take it from your post that you're not restricting the type of relationship to the romantic; if so, then The Fourth has characters of various species in various platonic relationships. There's also Unsounded, which has a significant difference in culture between the two protagonists.

First time using the forums here so please bare with me. I wanted to throw a recommendation out for a comic called morphE about 5 strangers who wake up in a mansion and are forced to learn magic under an over dramatic mage who wants everyone to like him, in spite of the whole kidnapping and killing thing.
It updates 3 times a week and reads like a visual novel.
(I realize this is a super old thread, but it is sticky. Also full disclosure, I'm a writer for the project)

I've been following Enemy Quest for a while now and it's got me hooked. The writing is sublime, particularly where characterization is concerned, and the plot keeps getting deeper and more complex, especially in regards to the main characters traumatic past.
Remarkably gripping, surprisingly dramatic and funny, and pretty NSFW at times. I highly recommend it.

That's a really broad (and a wee bit fiery) question.
If you're actually looking for a comic to read, try narrowing it down by listing your interests. Phrased that way, 9 out of 10 suggestions probably won't work for you, due to different tastes and such.

Hmm... If you like fantasy webcomics, two that come to mind for me are The Phoenix Requiem (which I believe is complete), and Unsounded (which is ongoing, I believe).
A word of warning, however: Unsounded can get into some dark and disturbing places at times.
My Games & WritingI think I would probably second Unsounded because it has the combination of wonderful art, story, and tone.
Then again, you could also say the same things of Paranatural, it just has a distinctly different art and tone. I think they're both amazing, but they aren't very similar to each other.
Had The Meek gotten further than three chapters, I probably would have put it in the same basket (but maybe slightly closer to Unsounded). It's resuming in a few weeks, I believe, so it may get there yet.
I guess those would be my 'greatest ever' picks. You could also make a case for Digger, Broodhollow, Lackadaisy or Order Of The Stick.

Looking at my list of webcomics, there are a few that I'm tempted to add, but which are either fairly early in their run (such as Harpy Gee), or are—for now, at least—orphaned (such as By Moon Alone).
In fact, there are quite a few webcomics that I'm tempted to list here, fairly new or not, each that I find wonderful in its own way. Without a more specific prompt it's difficult to narrow down the list, and I don't want to simply dump links to all of my favourite comics into the thread...

Quite an interesting story with endearing characters and tree puns all the way. I'm surprised we haven't made a page for it here!

Erstwhile has splendid art, and the storytelling is ... well, they do fairytales, so if you know most European fairytales there isn't much suspense, but there are nice little details such as names for the main characters and such, which you don't get in the usual fairy tale, so it is always interesting to see how the adaptation is.
So I've just been recommended The Property Of Hate by a friend, and I'm halfway through the archive and so far it's great. The art is lovely and it has some neat character designs. It's about a small child who is taken from her bedroom by a strange character who asks if she wants to be a hero.
