- Batman: The villain Two-Face was inspired by the Jekyll-and-Hyde story; the noble district attorney Harvey Dent has a psychotic break when half his face is scarred by a criminal, leading him to flip a coin to decide whether he's going to embrace his decent or darker impulses.
- Marvel Universe: Calvin Zabo, a Mad Scientist, is an in-universe fan of the book, made his own serum based on the one in the book, and even took up the name "Mr. Hyde" as a super villain alias. Unlike Henry Jekyll, though, who had a degree of shame, Zabo as himself is also a pretty horrible person.
- Doraemon: Nobita and The Space Heroes; There's a villain named Hyde who has a Meaningful Name. Like the novel's character, Hyde appears to be a reasonable and important figure of authority on the surface but is secretly evil on the inside and intends to eliminate an alien planet for his own benefit.
- Pagemaster: Richard Tyler, Adventure, Fantasy, and Horror visit Jekyll's and Hyde's house.
- The Deadly Experiments of Dr. Eeek: Jekyll and Hyde are mentioned early in the adventure; fittingly enough the book's main villain is another Mad Scientist, not unlike Jekyll.
- Jekyll: It's revealed that the novel is a trap to kill anyone seeking Jekyll's abilities, as the potion described is poison. The real source of the abilities turns out to be Jekyll's maid.
- Looney Tunes:
- Hyde and Go Tweet: Tweety ingests Dr. Jekyll's potion and turns into a gigantic canary monster to get even on Sylvester.
- Doctor Jerkyll's Hyde": A sequel to Tree for Two'', where Sylvester ingests the potion and attacks the bulldog Spike.
- "Hyde and Hare": Bugs Bunny is taken in by Jeckyll, then runs afoul of his alterego Hyde.
- "Hare Remover": Elmer Fudd tries to make a potion to turn Bugs into a "deviwish fiend". References the 1941 film version when, upon watching Elmer's reaction to the potion, Bugs tells the audience "I think Spencer Tracy did it better, don't you folks?"
