- Splinter's speech in the trailers for how family is important."You live...you die...you fight...as brothers. Remember, nothing is as strong as family."
- Raphael frantically rushing to the aid of the injured Splinter.
- Splinter thanking April for saving him and the turtles from the fire all those years ago and telling her that the care and love she and her father showed towards them in the lab (when neither human could have known that the animals were becoming intelligent) was what inspired Splinter to be a father figure to the turtles.
- While on the subject of Splinter, there's one that mixes with a Moment of Awesome. When the Shredder blitzkriegs in the sewers with a battalion of heavily armed goons and a titanic Powered Armor, a lone Splinter stares him down, katana in hand, and says "You will not touch my sons!" His whole battle against Shredder is not for vengeance, not for self-preservation, it is to protect his children.
- Raphael's blubbering speech in the climax."I just, uh… if this is our last moment together, I just want you guys to know I’m sorry! I’m sorry for being so hard on you! Every time I pushed you, I threatened you, I yelled at you, I pushed you beyond your limits is because I believe in you! I believe in each one of you! I believe in your spirit and your intelligence and your potential! And every time I talked about walking away, it was because I was SCARED! I just didn’t think I could stand next to you and call you brothers and say to you—I love you! I LOVE YOU GUYS SO MUCH!"
- The beat-boxing scene before the climax. Sure, it's a Funny Moment but there's more to this scene. Rather than just scolding Mikey for being silly in the heat of battle, the other three Turtles join in. You could view this as them realizing the possibility that this could be their last moment together and that they may not all come from the final battle alive, so they decide to have fun as brothers one last time.
- In most TMNT media, the boys refer to Splinter as "Master," "Sensei," or the still-somewhat-formal term "Father." Here? They call him "Dad."
- In the ending, Raphael thanks April for not exposing them to the world, and she replies "That's what family's for, Raph." He can only reply with an "Aw, shucks" face.
- Early in the movie, when April is unconscious after fainting at the sight of the Turtles, Donnie and Leo are clearly concerned about her possibly having a head injury from falling.
- They went out of their way to place her on something relatively comfortable before she woke up.
