• The various girls when hit by Dracula's Charm attack, all in dreamy voices.
  Charlotte: What a hunk~
  Yoko: Hello hottie~!
  Shanoa: Lord Dracula~
  Maria: Sooo dreaaamy~!
  • And also when it wears off and they're angry.
    Charlotte: What a jerk!
    Yoko: Son of a...
    Shanoa: [in a noticeably angry tone] Deceiver!
    Maria: Why you!
• As well as the various guys when hit by Astarte's Temptation.
  Soma: You are wonderful!
  Alucard: Such beauty...
  Jonathan: Your wish is my fantasy.
  Julius: My! What a woman!
  Richter: You're lovely!
  • And their reactions to it wearing off.
    Soma: What the hell am I doing!?
    Alucard: What!?
    Jonathan: Oh! Uh...sorry!
    Julius: Damn!
    Richter: What happened!?
• When Shanoa uses a teleporting portrait.
  (While spinning) I'm getting dizzy...
  • Heck, pretty much everyone’s reactions to the teleporting Portrait are hilarious! Some other highlights include Alucard sounding like a scared man on a rollercoaster, Soma suffers a voice-crack, and then there is Jonathan and Charlotte who just seem to be having a blast. Then again, they are from the videogame where travelling through portraits was a mechanic.
    Charlotte: Woohooooooo!
    Jonathan: Yahoo!
• Some of the guys' dialogues when you activate the "Oops" talk function are quite funny.
  Jonathan: Ugh, crap.
  Soma: CRAP.
• As in all Castlevania games, Lethal Joke Weapons abound, so it's always fun to kill enemies (or That One Boss) with them just to say that you did.
  • Case in point, defeating Menace with Paper Airplanes.
• Kicking butt with an 8-bit character (Simon or Fuuma).
• Charlotte when someone else gets charmed by Astarte.
  "You're dead!"
  • And its rarer variation...
    "You're ALL dead!"
• If someone you were playing with is using a DLC character you haven't unlocked/bought yet, they will show up on your screen as an Axe Armor with no remote attempt made to give the sprites any animation besides "walk" and "throw axe". The limited animation fudged to convey complicated things can be gut-bustingly hilarious for the person watching.
• Death's English laugh in his Chapter is hilarious.
  Death: "Hue HUE HUE HUE HEEHUEHUEHUEHEUEHAHAHAHAHAHAHAHAHA!"
