- Marth Debuted in "Smash Bros.":
- The casts of the Monster Hunter spin-offs, Monster Hunter Riders and Monster Hunter Explore make their Western debuts in this game, though in the case of Explore it did have a very brief test period in Canada.
- Otoranger was a mobile Rhythm Game based on Toku heroes, that lasted from 2013 to 2014, and never left Japan. Despite the short life span, over nearly 10 years later, the character Keyboard Mary appeared in the Turnabout Festival expansion, marking Otoranger's first official apperance overseas.
- Megami Meguri was a Nintendo 3DS game that ran from 2016 to 2020, and had a peripheral that was meant to work with Japanese train cards. In the supplementary cards for THE BEAUTIFUL 8 expansion, the main character Tsukomo was given her first official exposure to overseas fans.
- Meme Acknowledgement: The supplemental card "Wild Sparring Session" from the Schoolyard Royale expansion features Rajang giving a cheeky thumbs-up, a nod to the Japanese-originating meme of Rajang giving a thumbs-up with the caption "Ī-jan" (a pun on its Japanese name, "Rājan", with slang meaning "Ain't that good?").
- The Other Darrin:
- In English, Ashly Burch fills in for Laura Bailey as Chun-Li once again after Marvel vs. Capcom: Infinite. However, Bailey reprises her other role as Oichi, thus avoiding her talking to herself, in a similar vein with MvC:I, where she did not reprise Chun-Li, but reprised her other role as Black Widow.
- Hiryu is played by Yuji Ueda, his main voice actor in the Marvel vs. Capcom series. This is the first time he voices the character outside that series, replacing Hiryu's historical voice actor Kosuke Toriumi, who has voiced him in everything he has appeared in (beside the Vs games) ever since Strider 2.
- Role Reprise: Every character's voice actors from their series and crossover appearances return to voice their character here, in both English and Japanese.
- Ryu is played by Kyle Hebert in English, who's voiced him since Street Fighter IV, with the same applying to his Japanese voice actor, Hiroki Takahashi.
- Chun-Li is played by Ashly Burch in English, having replaced Laura Bailey in Marvel vs. Capcom: Infinite. Fumiko Orikasa voices her in Japanese since SFIV.
- Akuma is played by Richard Epcar, taking the role over from Dave Mallow since Street Fighter V. Taketora, his traditional Japanese voice actor since SFIV, returns.
- Morrigan is played by Siobhan Flynn in English, with Rie Tanaka voicing her in Japanese. Both have played the role since Marvel vs. Capcom 3.
- Jill is voiced by Patricia Ja Lee in English, who notably hasn't played the character since 2011's Resident Evil: The Mercenaries 3D.note Atsuko Yuya returns to play her in Japanese, having voiced the character in all video game and live action movie appearances since 2004's Resident Evil: Apocalypse.
- Wesker is played by D.C. Douglas, having been his voice actor since Resident Evil: The Umbrella Chronicles. Joji Nakata voices him in Japanese, who started voicing the character with MVC3.
- Ada is played by Courtenay Taylor in English and Junko Minagawa in Japanese. Both voiced her since Resident Evil: Operation Raccoon City, although Taylor was replaced by Jolene Andersen for Resident Evil 2 (Remake), with Teppen marking the first time she has reprised the role since then.
- X's English voice has Ted Sroka return to the role, who first played him in MVC:I. In Japanese, Takahiro Sakurai plays him, having voiced him since Mega Man X8.
- Zero's English voice is Johnny Yong Bosch, who played him since MVC3. Ryōtarō Okiayu plays him in Japanese, having been his sole Japanese voice actor since Mega Man X4.
- Dante is naturally played by Reuben Langdon, his voice actor since Devil May Cry 3: Dante's Awakening. Toshiyuki Morikawa plays him in Japanese, who began voicing the character with Devil May Cry: The Animated Series.
- Nero sees Johnny Yong Bosch playing him as per the norm since the character's debut in Devil May Cry 4, with Kaito Ishikawa playing him in Japanese since the Special Edition of said game.
- Oichi is played by Laura Bailey, her sole English voice actress since Sengoku Basara: Samurai Heroes. Mamiko Noto plays her in Japanese since the character's debut in Sengoku Basara 2.
- Sam Riegel returns to voice Phoenix Wright, having voiced him since MVC3. Takayuki Kondō plays him in Japanese, having begun with Phoenix Wright: Ace Attorney – Dual Destinies.
- Nina is voiced in Japanese by Kyoko Hikami, her original voice actress from Breath of Fire III. Her English voice actress is Abby Trott, this being Nina's first English-speaking role.
- Cammy's voiced by Caitlin Glass, her voice actor ever since Street Fighter IV, while her Japanese voice is provided by Miyuki Sawashiro, who has voiced her in all games from IV onwards as well.
- Like Dante, Vergil is voiced by Dan Southworth, his voice actor ever since his debut in Devil May Cry 3. In Japanese he's voiced by Hiroaki Hirata, who first voiced the character in Ultimate Marvel vs. Capcom 3.
- Ruiner Felicia is voiced by Gina Bowes in English and Kana Asumi in Japanese, both having voiced Felicia ever since Marvel vs. Capcom 3.
- Hiryu is voiced by T.J. Storm, who has voiced the character in both Ultimate Marvel vs. Capcom 3 and Marvel vs. Capcom: Infinite. In Japanese, he's voiced by Yuji Ueda, his voice actor in all Marvel vs. Capcom games (save Infinite, as it has no Japanese dub).
- Working Title: Known as "Project Battle" in the early alphas sent to prominent members of the Fighting Game Community.
