The surprise sixth member is the Beyonder himself
Which might make any physical conflict in the series a little less challenging.

Kid Loki will appear
He already made an appearance in Asgardians Of The Galaxy as some kind of corporeal spirit which was never fully explained, it would be pretty shocking if a Continuity Cavalcade book by Al Ewing built on Arc Welding and featuring a time-displaced Loki doesn't so much as mention that.

The real world is not beyond the House of Ideas
The One Above All mentioned that the House of Ideas is the end of the World of Action, and the beginning of the World of Creation. From those words, people believed that the World of Action is the comic book world, and the World of Creation is the real world where the comics are created.

However, that may not be the case. As the One Above All mentions this, we see a map that he painted, modeled after a Jacob's ladder of the Four Worlds in Kabbalistic philosophy. There are four sections of the map, painted in different colors: green, blue, yellow, and red. These colors represent a different world: Action, Creation, Formation, and Emanation, respectively.

With two other worlds beyond it, it's possible that the World of Creation was never intended to be the real world at all—it just happened to be a part of the Four Worlds that the Mystery was inspired by (with the possibility that the Enigma hailed from it).

Besides, the real world was already confirmed to be part of the multiverse as Earth-1218, so that alone should debunk the "real world is the World of Creation" rumor.

Yahweh from Ghost Rider and Howard the Duck MAX (if it's canon), as well as the Fulcrum are two of the "hands" that are the One Above All but also themselves.
The former, to explain why there are seemingly two monotheistic Gods, and the latter because it's a very popular fan theory with a lot of evidence to back it up.

Jason Quantrell was secretly training Elsa Bloodstone to defeat The Enigma.
Elsa is Marvel's biggest Continuity Snarl right now, and she was a member of Nextwave so she's already been a victim of The Beyond Corporations reality warping. Maybe all the changes are the result of "Quantrell" periodically altering her history trying to create a version of her that's strong enough to defeat the Beyonders' most feared enemy.

The White Hot Room and the Land of Couldn't-Be-Shouldn't-Be have opposite counterparts
As seen in Al Ewing's other comics, both the Beyond and the House of Ideas both have locations considered to be their opposite counterparts: the Un-Beyond and the Below-Place, respectively. If so, then it only makes sense that the other places in the Mystery that we've explored so far have their own dark counterparts.

The Train and its Conductor from Defenders: The Best Defense hails from the Mystery, particularly outside the House of Ideas
Galactus mentioned that the mysterious Train and Conductor hail from a space and time beyond his understanding. Perhaps that's foreshadowing that they both hail from outside of space and time. And knowing Al Ewing's love for Arc Welding, it's possible he'll address the Train's true origins at some point.

The World of Creation is part of the megaverse
The House of Ideas is said to be the furthest point of the entire multiverse. So what lies beyond the One Above All's domain? One possibility is that there's a megaverse that houses not only the multiverse, but all multiverses. For example, the Megaverse can house the Avengers' multiverse, as well as the Justice League's multiverse (the crossover JLA/Avengers outright confirmed that the two multiverses were part of the megaverse), as well as an infinite amount of other multiverses, each with their own unique set of heroes and villains. And the World of Creation could be a hub for every multiverse, and the home of its creators.

There will be a limited series focusing on the Beyonder in the future
At the end of Defenders: Beyond, the Beyonder decided to leave the multiverse for good. Perhaps this could lead into a miniseries focusing on the Beyonder's adventures outside of the multiverse. It could allow us to catch a glimpse of what lies beyond the House of Ideas, and even tell us a little more about the Enigma.

There's a World of Destruction opposite the World of Creation
The One Above All mentions that beyond the House of Ideas, and the World of Action as a whole, is the World of Creation. But much like how places like the Beyond and the House of Ideas have dark counterparts of their own, the World of Creation could also have its own counterpart. And since the opposite of creation is destruction, there could very well be a World of Destruction beneath the Below-Place, with the One Below All being the start of something even lower.
