asdfgl Shin Megami Tensei! I've said this for years. The series has a hugely deep character creation/customization system, but once you beat whatever uberboss is waiting for you, there's no point to it anymore. Player vs Player combat would add a whole new layer to the game.
Granted, abilities and moves would need a bit of an overhaul. Can't have people casting Mediarahan every turn and expect the game to stay fun. You'd also need to adjust abilities that grant elemental Absorb/Reflect/Null, perhaps by having them grant another weakness when used? That way you could keep the enemy guessing about your weaknesses. It'd still be possible to make a no-weaknesses critter by chaining elemental resist abilities together, but it would take a lot of moveslots. And I think you'd probably have to reduce the amount of HP physical skills take to use, or switch them over to MP like Strange Journey did.
But anyway, the series has a lot of potential in this direction, so I'm kind of shocked it hasn't been done already.
But somehow,◊Deus Ex. I don't mean a typical deathmatch thing or randomly generated survival mode, I mean a dedicated, quality, handcrafted coop campaign. Replay value wouldn't be the primary attraction, instead, all of the puzzles and such would be designed specifically around requiring multiple players to do different stuff in a coordinated fashion. Crucially, it would keep Deus Ex's stipulation that there be multiple equally valid ways of beating a level, though in this case each would also require all the players' cooperation.
The Dragon Age and Mass Effect series could benefit greatly from pick-up co-op, either multiplayer or splitscreen. Actually, just splitscreen; the tactical pausing in those series' gameplay would wreck merry hell on connections.

I'd like to see it added to The Legend Of Zelda. Four Swords Adventures was a good example of how multiplayer might work, but it was pretty flawed. The levels were long, making each level a long commitment that all players have to agree to ahead of time. Having 4 copies of Link at all times was rather awkward, as even in single player you had all these other characters following you in a straight line or walking in formation, and it was annoying to look at. And finally, requiring each player to have a GBA and a GBA-GC link cable was just plain stupid.
But that idea could be done again, or maybe an online multiplayer Zelda instead.
The basis is there. It could be level-based, just like Four Swords Adventures. Collecting rupees or some other item would provide a reason for competition, while players would still have to work together to solve puzzles or fight enemies as a group. Maybe have checkpoints and a bigger penalty for death (Four Swords Adventures had you collect fairies, which were so plentiful that you had virtually unlimited lives anyway). If all players die, you revive at the checkpoint, New Super Mario Bros Wii-stylle.
Randomized elements in levels, kind of like what Left 4 Dead does, would also help prevent things from getting boring and redundant. Finally, allow players to create their own character. Do we need 4 Links? Or even 2? How about, say, a hardened elderly warrior, or whoever someone wants to create? Twilight Princess hinted at Link having friends who help out, yet you don't see them helping out in battle. Create-a-character would get around that nicely.
Anyway, that's how I'd like multiplayer to work in Zelda. What games do you think could have a great multiplayer, and also, how would you design it if you had the chance?
