- In PreCure in a Nutshell, Narcistoru gets the parody name of Narcis Prince.
- The Simpsons: Team L.A.S.H. contains a scene where Mr. Burns is helping his adoptive daughter Anastasia train for a gymnastics meet. One of the things Mr. Burns does is have her chase after a bicycle that he is riding, in an obvious allusion to the famous "Little Mac trains with Doc Louis" scene from Mike Tyson's Punch-Out.
- In the Strong and Steady story "Queen Bee Tongus
", Tongus names one of her roses Flamenco, after Don Flamenco.
- The Super Mario Bros. Movie: Early in the film, Mario and Luigi are eating at a Brooklyn restaurant called Punch-Out Pizzeria. Little Mac, Doc Louis, and many of the opponents appear as photographs hanging on the wall.
- Henk Rogers mentions this game in Tetris (2023).

- In The Lonely Island song "Incredibad", the trio are said to be playing Punch-Out at the beginning.
- The Goldbergs: A Punch-Out arcade machine appears in Age Of Darkness.
- WarioWare games often include a Punch-Out microgame in the 9-Volt levels.
- Deltarune: The second chapter has a moment where Queen challenges Kris to a Punch-Out-like arcade game. The Final Boss fight against GIGA Queen plays the same way, complete with Non-Standard Game Over screen that parodies the Game Over screen from Super Punch-Out.
- Ms. 'Splosion Man: The boss fight against the Bride plays out just like a match from Punch-Out, complete with intermissions that parody the same ones from the NES game.
- One of the retro games that Joker can play in Persona 5 is “Punch Ouch”, which has a title screen, music, and sound effects that are similar to the NES game. This reference would come full circle in Super Smash Bros. Ultimate, as Joker and Little Mac can fight each other.
- In Fans! Marc is challenged to beat Mike Tyson in Punch-Out. Having not heard of the game he attacks the real Tyson. While recovering in the hospital afterward he tries the game to see how it compares.
- In Phelous's review of "Animal Soccer World", Phelous adds the main Punch-Out theme to a scene of a panther and a dog fighting.
- Half-Life VR but the AI is Self-Aware: Dr. Coomer states that if he could live in any video game, it would be Super Punch-Out for the SNES, a favorite from his youth. It's fitting for his love of boxing, and seems to be his way of asking Gordon what's going to happen when the game— and their world— ends.
- Family Guy:
- In "Not All Dogs Go to Heaven", Meg makes the family say grace at the dinner table, and Peter prays for God to give him cheat codes for Punch-Out, saying that he's been stuck on Bald Bull for years.
- In "A Fistful of Meg", Quagmire and Meg reenact the famous "Doc Louis training Little Mac" scene, with Quagmire riding a bike and Meg running after him. There's even an Art Shift to NES-style graphics.
- Muppet Babies (2018): Fozzie's Training Montage in "The Karate Cub" features a scene where Fozzie chases after Piggy riding a bicycle similar to the montage in Punch-Out!!. The scene is even animated with 8-bit sprites.
- Robot Chicken: A sketch from "Up, Up, and Buffet" features Doc Louis dying from a heart attack during the training segment, and at his wake, several of Little Mac's opponents give personal eulogies.
- The Simpsons: The subplot for "Moaning Lisa" involves Homer training to beat Bart at Super Slugfest a parody of the Mike Tyson version of Punch-Out.
