I believe it's basically when a certain quirk, or set of quirks for which a character is known show up in their relatives too, so it's implied that said quirk(s) are hereditary. For instance, if you've got a character who happens to be a Pyromaniac Fighting Irish, and we get to meet their Irish family, it turns out they all have a perchant for violence and a knack for Stuff Blowing Up. The mom, the dad, the cousins, and their Famous Ancestor are all pretty much like that. Because well, it runs in the family.

Are the Winchesters really a good example of this? There aren't any "normal" family members, and they are perfectly aware of how strange they are.
