• This line about Martha on the 5th floor of the tower, said in complete deadpan:

  Luke: I guess she followed us in here.

• Messing around with the furniture in the Inn can yield some hilarious results. For example, putting both beds into the same room will put one on top of the other like a hastily-made bunk bed, with Luke commenting on it.
• At the end of the game, Layton asks Luke to find a secret hidden on the painting of Flora in the Reinhold Manor. Touch the painting's cleavage, and Layton will bemoan Luke for acting ungentlemanly:

  Layton: Now, Luke, it's important to be a gentleman.

• Layton loves his car, but Luke sees it for the old relic it is, and his Innocently Insensitive comment on how no one would want to steal it leads to one of the rare times Layton is either oblivious, or offended:

  Layton: There's the Laytonmobile. It seems unharmed.
  Luke: Don't worry, professor. No one would dream of stealing the Laytonmobile.
  Layton: ...Just what do you mean by that, Luke?

• Luke messing around with Don Paolo's Chelmey mask in the credits, making Flora laugh out loud.
• Also in the credits, Flora is gleefully cooking a meal, but said meal looks deathly atrocious, to Layton and Luke's horror.
