- The Original Darrin: Leon and Chris are voiced once again by Matthew Mercer and Kevin Dorman respectively, marking Role Reprises from Resident Evil: Vendetta and previous worksnote . They in turn ended up replacing Nick Apostolides (Leon's voice actor from the remakes of 2 and 4 in addition to Infinite Darkness) and Jeff Schine (Chris' voice actor from Village). In addition, Salli Saffioti returns as Ingrid Hunnigan after being replaced by Raylene Harewood in the remake of 4.
- Role Reprise: Erin Cahill and Cristina Valenzuela return to voice Rebecca and Maria respectively five years after Vendetta. Stephanie Panisello and Nicole Tompkins also return to voice as Claire and Jill respectively.
