*** Alucard directly states that the only way to kill Anderson (and himself as well) is to remove the heart. In both the original manga and the OVA, Alucard blasts away at Anderson but can do no more damage to him as his body continuously regenerates. In the manga's case, this is more along the lines of Alucard trying to slowly blow Anderson's defenses away so he had a spot to reach the heart. Alucard's attack with his final bullet ultimately proves futile when Anderson's vines stop the bullet from reaching his heart. Therefore, this in itself is not a plothole.
*** However what can be described as potential plot hole is the fact that whenever manga Anderson utilized his vines, any and everything they touched instantly went aflame and turned to dust. He destroys the rest of the Level Zero army this way and it most likely because of this that Alucard never directly attempts to fight Anderson, instead pushing his familiars in front until he has an opening to take Anderson out. This is the exact opposite in the OVA where Alucard directly interacts and rips through the vines.

In the manga this inconsistency could be explained by the fact that Casull inflicted the head shot, while his legs were shot off by Jackal. In the OVA, Jackal was the cause of all three wounds.
