- All-Star Cast: The voice cast is pretty big, especially compared to other animated series. It includes Kaley Cuoco, Lake Bell, Diedrich Bader, Alan Tudyk, Sanaa Lathan, Ron Funches, J.B. Smoove, Jason Alexander, Wanda Sykes, Giancarlo Esposito, Natalie Morales, Jim Rash, Tony Hale, Christopher Meloni and Rahul Kohli.
- Billing Displacement: By season 2 it's clear the show has two main protagonists, Harley and Ivy, with both of their actors getting about as many spoken lines. However, in the end credits only Kaley Cuoco has a full credit card for herself, whereas Lake Bell is listed among the supporting actors, probably because she's much less famous than Cuoco.
- Channel Hop: Seasons 1-2 were made for DC Universe. Starting with season 3, the show moved to HBO Max, due to DC Universe being rebranded as a digital comics app.
- Children Voicing Children: Damian Wayne is voiced by a kid, Jacob Tremblay, rather than an adult woman which is usually the norm for pre-pubescent boys in animation.
- Creator's Favorite Episode: Series director Colin Heck says that "There's No Place To Go But Down" as his favorite episode, particularly with Harley and Ivy's kiss at the end.
- Cross-Dressing Voices:
- "Til Death Do Us Part" has Joey voiced by Vanessa Marshall.
- The episode "A High Bar" has Marcus and Benjamin voiced by Meryl Hathaway and Nicole Sullivan.
- Descended Creator: In addition to voicing Harley, Kaley Cuoco is also an executive producer for the series.
- Executive Veto: Although the team behind the show has said that the people at DC are very supportive and willing to let them do what they want, a scene where Batman preformed oral sex on Catwoman was cut out of season 3. The reasoning is that “heroes don’t do that” (in context, the quote is talking about how making a show about villains frees them up from content restraints) and Batman sells too many toys even though the show isn’t targeted at kids.
- In Memoriam: The Season 4 finale episode opens with a dedication to Arleen Sorkin, the original voice and inspiration for Harley in Batman: The Animated Series, who died midway through the season's airing.IN MEMORY OF ARLEEN SORKIN 1955-2023
- I Want You to Meet an Old Friend of Mine:
- The show also brings Newman and George Costanza as the Penguin and Sy Borgman, respectively.
- Harley and her father have the same voice actors as Brandy & Mr. Whiskers and as Weathervane and Optimatus.
- Non-Singing Voice: Jonah Platt sings for Clayface in the season 2 finale instead of Alan Tudyk.
- The Other Darrin: In Season 4, Zeno Robinson replaces Scott Porter as The Flash.
- Permanent Placeholder: Charlie Adler revealed that he wasn't the first choice to play Nick Quinzel. A famous actor was considered for the role while he did temp dialogue, but Dean Lorey liked his performance and he was cast in the role proper.
- Playing Against Type:
- Charlie Adler is best known for voicing Comedic Relief Characters who are either Nice Guys or villains that are too silly to take seriously. Nick Quinzel, however, has no likable traits whatsoever and only a select few of his moments are Played for Laughs.
- Tony Hale’s two most famous roles have him as one of the most likable characters in a group of mostly unlikable people. Here, he’s the Token Evil Teammate, to such an extreme degree that he becomes the main villain of the second season.
- Harvey Guillén, a Camp Gay comedic actor, voices The Comically Serious Nightwing, and he does so in a gruff voice that's all but unrecognizable. But it's implied that Dick is purposely deepening his voice to sound more intimidating, as his voice is much higher when he's upset, at which point it sounds closer to Guillen's.
- Playing with Character Type: Michael Ironside reprises his role as Darkseid from the DCAU, though his depiction here is slightly sillier than his other roles.
- Production Posse:
- The show reunites the showrunners of Powerless, Justin Halpern, Patrick Schumacker and Dean Lorey, with two of its lead actors, Alan Tudyk (Van) and Ron Funches (Ron).
- Harley and Ivy's Season 4 future daughter is voiced by Kaley Cuoco's The Flight Attendant co-star, Zosia Mamet.
- Real-Life Relative: Kaley Cuoco (Harley) and Briana Cuoco (Batgirl) are sisters in real life.
- Release Date Change: Season 2 was originally going to come out in fall 2020, but was moved to April 3.
- Role Reprise:
- Diedrich Bader reprises his role as Batman from Batman: The Brave and the Bold.

- What Could Have Been:
- Originally, Harley's DC Extended Universe actress Margot Robbie was on top of the wishlist to voice her here. For unknown reasons, this never came to be, and Kaley Cuoco voiced her instead.
- Being Harley Quinn was originally going to show many versions of young Harley before eventually settling on one version.
- According to an interview conducted with the show's creators by Variety, they originally planned to have a sex scene between Batman and Catwoman in season 3. DC rejected their proposal, telling them that "heroes don't do that" and that "It’s hard to sell a toy if Batman is also going down on someone.".
- According to an Entertainment Weekly interview, Justin Halpern considered retconning Bane's stupidity compared to his classic genius portrayals as Obfuscating Stupidity as part of a larger mastermind plan, but this idea was quickly shot down.
- Write What You Know: The spin off comic "The Eat. Bang! Kill. Tour" has a plethora of queer black women. The author Tee Franklin is self described as a "black, queer, autistic, disabled comic writer".
