When in doubt? Throw the book at them!note

It wouldn't be Rise of the Teenage Mutant Ninja Turtles without a healthy dose of comedy, would it?

  • The fact that right after the Downer Beginning of Casey fleeing the Bad Future and Future Leo and Mikey sacrificing themsevlves to help him escape, we cut to the Turtles themselves... with Leo balancing on a bridge with a ton of pizza boxes stacked on his head, trying to break Donnie's record and successfully doing so, much to Donnie's dismay. Mikey and Leo urging him to cheer for this moment and Donnie's response sells it.
Donnie: (in an utterly deadpan voice) You, sir, have bested me. Mazel.
Mikey and Leo: Ahem. Ahem, ahem.
    Donnie: Leo. Leo. Leo.
  • While chiding Leo on his immature attitude, turns out Raph is still quite self-conscious of himself, with Leo quite well aware of it.
Raph: What's it gonna take for you to be serious, Leo?
Leo: And end up with a worry line between my eyes like you? No, no, no, I do not want a Raph chasm.
Raph: Chasm!? (he starts to feel his eyebrows nervously)
Leo: You know, valley, gorge, crevasse...
    • And then when Raph goes on a tirade explaining that he's responsible for looking after his younger brothers, turns out Leo wasn't listening to any of it, reading one of his comics. It's enough to spur Raph into trying to attack, with both Donnie and Mikey frantically trying to keep Raph and Leo from fighting, and when Splinter intervenes to tell them to stop fighting? Turns out that alongside them being family, he also wants them to stop because they're interrupting a TV drama he's watching.
  • When Casey is first explaining that he is from the future he explains how humanity was hunted and forced to eat leaves and rats to survive, before telling Splinter no offense. Splinter's response? "None taken. We are delicious." This becomes a Brick Joke at the end of the film when Casey tries pizza for the first time, happily declaring it way better than rats, with Splinter being disappointed upon hearing this.
  • When Warren and Hypno-Potamus see the Krang finally arriving from the prison dimension, they realize how utterly and completely out of their element they are, and with a frightful, "Ooooh..." slide back into the shadows and are never seen for the rest of the movie.
  • When he, Leo, Casey, and Mikey are trying to find Raph, Donnie lets slip that he secretly implanted a tracker on Raph, and Leo asks if that means that he put trackers on the rest of them as well. Donnie's denials are far from convincing.
Donnie: Nooo...? Nooo... Of course I did... n't.
  • Among the many attempts April and Splinter pulled to break the Krang's key, they tried microwaving it, flushing it down a toilet, eating it, and using a Zettaflare.
    • Also included on their list? "Bean fight" and "wet punch". So is spanking.
  • During the movie's Darkest Hour, we get this exchange between the heroes:
Casey Jones: Guys, it's time to face the facts. We can't change the future.
    Mike: Says who?
Don: Says the Grandfather Paradox, says physical determinism, says the space-time continuum. Look I didn't want it to be true, but… (sighs) science.
    Leo: That's it! Donnie's wrong!
Don: The world is ending, yet the barbs continue.
  • While going to attempt to hijack the Technodrome, Donnie and Mikey both come across an odd mass of Krang-y flesh serving as the main systems, much to their confusion. As they try to figure out how to use it... Donnie's face turns into one of absolute dread as he realizes he needs to merge with it.
Donnie: Oh God. I know what I have to do.
    • He and Mikey (upon realizing as well) struggle to avoid vomiting at the idea of it, with Mikey trying his best to encourage Donnie to go through with it while retching the whole way.
Donnie: [retching and nearly vomiting] No. No, I don't think I can do this!
Mikey: What are you talking about? [he looks back at the Krang-y flesh system] Oh no, that's... [Mikey begins retching as well] so gross!
      Donnie: I can't do it!
Mikey: [shaking his head] I believe in you!
Donnie: Augh, it's my worst nightmare!
Mikey: [still trying not to retch] I know!
Donnie: I'm doing it! I'm doing it!
[Donnie looks away, closes his eyes and thrusts his hands into the Krang-y flesh system, Mikey covering his mouth as he does]
  • Splinter (and basically all the viewers watching) becoming sick of Krang Two's constant desire to kill something, asking her to demonstrate some other trait.
    Splinter: Where's the Character Development?
  • At the end, when Casey is looking at the group photo they all took on April's phone, he notices she got a text... from someone named Cassandra. April opens the text to see Cassandra Jones took photos of herself kicking all kinds of Krang butt, all while wearing a very familiar hockey mask. Cue a mortified Casey; "THAT'S MY MOM!"
Donnie: There goes the space-time continuum...
