Lou is hiding some heavy flaws.
The trailers call him the "most perfect doll" of Perfection. If he's so perfect, why is he the leader of Perfection that teaches the newcomers to be perfect for the real world, and not in the real world given to his own child? He berates the Uglydolls that are curious of Perfection for "not being perfect". This could possibly be his own flaw, which he is secretly hiding, which will be a huge game-changer in the movie's plot.
  • Confirmed in away. Lou is actually a prototype Perfect Doll that was never meant to actually be given to a child.

Mandy either was or will become, an Uglydoll.
As the most prominent Perfection doll that seems to support the Uglydolls and their plight, she could also be the one that embraces her flaws the most. This could lead to her becoming an Uglydoll as a badge of her differences, or she already was an Uglydoll, who tried to cover up her flaws due to personal shame of them, becoming a Perfection doll in the process. Either way, she shares the warm hair/fur color, hair "bun", and a similar name with a canon Uglydoll that is still in the movie's toyline, despite not showing up so far in the commercials: Mynus.

Jeero and Big Toe are related familially.
Am I the only one who thinks they look similar? I don't think it's too farfetched to believe that they're related somehow, just like how Wage and Abima are cousins.

Nolan's imperfections were created through him being scanned from the broken scanner.
The same scanner that Moxy went through. During the repair, it wasn't entirely fixed, leading to some mishaps, which led to it going wonky and giving Nolan his unique design.

Ugly Dolls and Toy Story take place in the same universe.
The pre-Big World training is what instills in most toys that the most important thing in life is to make a human child happy. The Buzz Lightyear franchise toys, due to a glitch, get actual space ranger training (or whatever role in that storyline is appropriate to them, such as the Zurg toys getting Evil Emperor training), which is why they don't initially realize they are toys. This is also why Forky, in the fourth Toy Story movie, doesn't initially understand what being a toy is about - as an improvised toy made by a child, he didn't get this training.
