     General 
  • The improved facial capture engine within the game allows for more expressive and unsettling faces on the character models. Special mentions go to the Yamibito as they evolve, Yuri Kishida when she shows a face on her chest, Simulacrum Ichiko upon going berserk, and the monstrous Otoshigo.
  • In some of the archives, you can find a magazine about paranormal activities, dedicated to a strange creature living on the island no one has ever seen, and a video showing evidence of the existence of that creature (like Big Foot, or Nessie, to put it in context). Now, in a late mission in the game, you can Sightjack with the Shibitos... and something else. And that something is real, and is out there, lurking in the shadows.
  • The Shibito in Siren 2 now resemble shambling corpses that are inhabited by ghostly smoke that invade their bodies, as opposed to the more lucid and indoctrinated variants from Siren 1.
  • The Yamibito are essentially stronger, faster, and smarter variants of the Shibito, and they are easily identified as pale humanoid beings cloaked in black fabric. They behave and speak more coherently than the more zombie-like Shibito and even express utter glee in hunting down their prey.
    • And upon evolving? Their heads enlarge to such a grotesque size, and either resemble a crawling critter with bird-like talons, or a torso resting onto their giant head that walks with their enlarged fingers. Unlike the aquatic and insectoid-themed Shibito in Siren 1, the Yamibito's evolution is outright bizarre and nonsensical.

     Story Events 
  • Yuri Kishida lures Mamoru Itsuki to the Underworld and unbuttons her top to show her horrible secret: she is a 'dove', an offspring of an ancient being known as Mother, whose face emerges from Yuri's chest. Mamoru could only stare in horror while a headless mermaid-like creature erupts from the ground to merge with Yuri's body, now that she has completed her mission.
  • Shu Mikami is lured to his demise by a vision of his beloved Kanae in the Underworld, who embraces him before he is devoured by Mother.

     Memorable Stages 
  • The abandoned theme park on Mamoru's third level begins with a mission objective unlike any other: open the gates to the Underworld. There has been no mention of the Underworld up to this point, and Yuri Kishida is adamant about reuniting with her 'Mother', who is being held captive somewhere. She then instructs the player to guide her to all seven gates scattered across the level and unlock them, where a gaping hole emerges from the ground and reveals a hellish landscape underneath. Mamoru's desire to help Yuri reunite with her 'Mother' ends up inadvertently awakening an ancient and hostile being, who gives birth to a herd of Yamirei in the real world, and he never realizes his error until it's too late.
  • Yorito Nagai's third level seems like your standard rescue mission, as the player is expected to reunite with Ichiko Yagura who is hiding somewhere after being separated. Upon meeting her, however, she suddenly goes insane and starts gunning down anyone standing in her way, all while babbling about being reunited with Mother. It's at this point that the player will learn that the Ichiko Yagura we've known was in fact dead all along and was replaced by a 'Simulacrum', a crude replica of the drowned girl.

     Unique Shibito and Yamibito Characters 
  • The fate of the real Ichiko Yagura, who had drowned years ago in the past after having fallen overboard from a ferry she was on. Her corpse was found by an ancient being named Otoshigo, who then created a replica of the drowned girl known as a 'Simulacrum'. The Simulacrum Ichiko then wakes up in the present time, believing itself to be the real Ichiko that had been brought back on the ferry. As time passes on, Otoshigo gradually takes over Simulacrum Ichiko's mind, and eventually goes on a rampage, killing anyone who stands in its way in order to be reunited with Mother again. After having failed to meet Mother once more and having fallen off the pylon tower, Simulacrum Ichiko eventually dies from its wounds, thus angering Otoshigo and prompting the ancient being to resurface and lay waste on whoever stands in its way.
  • The second final boss from Yorito's final level introduces Otoshigo, a gigantic, malformed, and rotting head covered in a mass of arms and hands.

    Others 
  • After Siren 1, there was the print ad for Siren 2. A close up of a woman's face. With the eyelids sealed shut. That brought back uncomfortable memories of the trailer, as well as being terrifying.
