The main form of Villainous Incest to me is the Milfy Wicked Queen-Mother or High Priestess seducing her puppet Prince or Boy King who is her Son. The key example of this coming from real like Agrippina and Nero. SO I'm surprised their not mentioned here, either there of via fictional depictions of it. (I, Claudius and A.D. both depict it in disappointing ways).
Other examples could arguably include Alexander and Olympias, Semiramis, some interpretations of Isis and Horus or Jessica and Paul Atredes or even Cersie and Jofori on Game of Thrones.

Removing the Preacher mention because there is nothing villainous about it.
I'm not sure whether or not to add Illumi Zoldyck from Hunter x Hunter. He starts off as a Lawful Evil character who gets progressively creeper & more insane as the series goes. He canonically has a "twisted love" for his little brother, but he is not outright stated to be IN love with his brother. Killua is terrified of him & reacts around him as if he had been sexually abused, but it's more of a "mind rape." He is actually rather comparable to Seimei from Loveless.
