I would say likely. While normally we assume the creator's intent based on content to judge whether tropes fit, Designated Villain is when the creator's intent is directly contrary to what appears in the work.

I don't really have one. I got the dilemma while reading Of Vale Blood, but while there is some serious Designated Villainy there, it stems from different reasons.
Apathy is Death. Worse than Death, because at least a rotting corpse feeds beasts and insects.

If a character exibits Adaptational Heroism, but the story tries to paint him as just as evil as he is in the original, does that count as an example of the trope?
