Average student Tsukune Aono does not get accepted into any local high schools but his parents are more than happy to have him enroll in the boarding school Yokai Academy after his father had a lucky encounter with a drunk man in a white monks robe. He encounters and befriends Moka Akashiya, a vampire that becomes enchanted by the scent of his human blood. However, he discovers that Yokai is a school for monsters. According to the school rules, the students must retain their human forms on academy grounds and any humans who manage to get past the schools magic protection barrier are to be executed. Tsukune tries to run away from the school but has a confrontation with Moka. A fight with a delinquent student named Saizo Komiya quickly endangers Tsukune's life right before he accidentally removes the rosary on Moka's neck, which transforms Moka into a powerful vampire with silver hair that easily defeats Saizo. Afterwards, he wakes to find the pink-haired Moka caring for him and wanting another drink from his neck.

** For Tsukune, it's after he realizes that he is in a school full of monsters. When it has become clear that Outer Moka (whom every guy is instantly smitten with) has affections for Tsukune, every guy in the student body gives Tsukune the stink-eye and Saizo to antagonize him instantly, only for Moka to defend him. This establishes him as a CluelessChickMagnet and an UnluckyEverydude who is completely outmatched by his monster classmates, but he is such a NiceGuy that the girls that flock to him are ready to fight on his defense.
** Outer Moka meets Tsukune by crashing her bike into him, having become light headed from anemia. She overlooks that his hand is on her leg and grabs a hanker-chief to help when his nose bleeds, only for her to bite his neck when the smell of his blood is too much for her. This establishes her as being sweet and a little clumsy, with her more monstrous traits presented as harmless and endearing.

* FantasticRacism: Monsters have a general distate towards humans (see FelonyMisdemeanor for context), but Moka dislikes them because she was routinely bullied at humans schools her whole life. It is not until she gets to know Tsukune better [[FormerBigot does this opinion change]].
* FelonyMisdemeanor: It is established that while Yokai Academy was founded to help acclimate monsters to live among humans, any human that were to find their way into the school's borders (by accident or intentionally) would be executed on the spot. This encourages Tsukune to try and leave, only to decide to stay and pretend to be a monster at the end of the episode when Moka promises to keep it a secret (and because the bus won't return for another month).
* LecherousLicking: Saizo is seen doing this at the thought of [[ToServeMan eating humans]] and at the sight of Moka.
* MeetCute: Tsukune meets Moka when she crashes her bike into him when he gets lost in the woods.
* MonsterOfTheWeek: Saizo is an [[OurOrcsAreDifferent orc]] with a contempt for the idea of coexisting with humans and does little to hide his lust towards Moka. He attacks Tsukune for hanging out with her and later corners Moka with the intent of "claiming" her.
* PowerLimiter: It is established that Moka's rosario acts as this. Tsukune is the only one who can take it off, and when it does, the all-powerful Inner Moka is released.
