I could mention Gary Owens, who, while normally known for his comedic roles - such as Roger Ramjet and Powdered Toastman - actually managed to play the original Space Ghost rather well.

If you've seen the film Shakes The Clown, he plays the villain, and manages to actually be a pretty convincing villain in some parts of the film.

Stretching the definition of "serious" a bit, but how about Brian Cummings as the announcer on a season of Let's Make a Deal in the 80's?
Tom Kenny can do serious pretty well. His Doc Ock in Ultimate Spider-Man was played pretty straight considering the show is mostly comedic. He also surprised me as Deadshot in Brave and the Bold.
Even though the show is mainly an adult comedy, Will Arnett hit some pretty serious moments at times in Bo Jack Horseman as the title character.

Would Adam West count? If so, he had some good serious moments as Batman on the last two seasons of Superfriends (yes, the show wasnt the most serious show out there, but he played the role straight) as well as The Gray Ghost on Batman The Animated Series.

That he is indeed, old chum, but ever since Batman ended it's generally been as a source of comedy.

This thread is for examples of voice actors mostly known for comedic and cartoony roles playing characters that are serious, creepy or just plain heartwarming.
A perfect example of this is Grey De Lisle. I first knew her as Vicky, the evil babysitter from Fairly Oddparents. Her love of torturing Timmy Turner was always played for laughs. Then I saw the second season of Avatar The Last Airbender where she voiced Zuko's sister, Azula. While this character also loved to take pleasure in other people's pain, there was something legitimately unsettling to how she was portrayed.
Another example that comes to mind is the character Fixit from the Teen Titans episode "The Sum of His Parts". He was played by Tom Kenny, known for wacky roles like Spongebob Squarepants and another character on Teen Titans, the villainous magician Mumbo Jumbo. Here, he plays Fixit with almost no emotion to reflect how much of a machine the character has become. It's very reminiscent of Michael Ansara's Mr. Freeze from Batman The Animated Series.
