I'd say games that are meant to be beat in one go (Bad Dudes Vs Dragon Ninja and Aladdin which don't even have a Password Save, and arguably Portal) shouldn't really count.
But outside of this type of games, the fastest I've ever beat one was The Legend Of Zelda: A Link To The Past, and my record is two days, not one.

Back in the day when I still had it, I could beat Megaman X4 in one sitting in like an hour or two.
Slightly crazier: Tales of Vesperia, for the speed-run achievement that requires you to beat the game in under 15 hours.

I would've beaten Halo ODST in a single day as well but I got caught up in doing the sidequests about the city monitor and getting some of the achievements, so it ended up taking me three days on that one.
I beat Turok (N64) in 5 hours, then I saw someone beat it in 40 minutes
If it makes you feel any better, that was probably a TAS. The fastest verified speedrun of Turok is a little over an hour.
On topic. Most recently, I've done this with Portal and Batman Arkham Asylum. Portal was a second run, though.

It's not exactly naive. And it can happen. But it's tough. And definetly worthwhile.The Smurfs on the SNES had a Password Save, but the last Act turned out to be so frustratingly difficult that starting there was a guaranteed Game Over. So the best strategy was to play it entirely in one go, collecting 1-Ups along the way to spend them all in the last Act.

Mario Kart: Double Dash took me a single day to beat all the Circuits.
I did beat Quest 64 in one sitting, technically. I had to leave the n64 on for 3 days, but I had school and such, and no memory card. So it would've probably taken me a single day at most. On the other hand, it was my first time playing through, so I took my time on it.
