Cynthia Utrom will indeed be an Utrom linked to the turtles’ origins.
Obviously she’s going to be that. They’re giving it away in the name. Most likely she will be a prominent scientist or tech mogul. The only question is, will she be friend or foe.
  • Or alternatively, she'll be revealed to be Krang in disguise.
  • Her real name will be Ch'rell.

Splinter will be Hamato Yoshi.
The nature of this film seems goofy enough that the sensei will have once been human.
  • Jossed, a new clip revealed, indicates that it is not the same case, since the clip that reveals the origin of Splinter and the Turtles a bit, Splinter was an ordinary rat and was never a human.

Shredder will be teased in a post credits sequence.
Confirmed.

Bebop and Rocksteady will be hired by Shredder for a Sequel Hook.
With the recent confirmation that Bebop and Rocksteady will be in the film, there may be speculation that they will at some point be recruited by their future boss Shredder, for the two of them to be Shredder's loyal henchmen.
  • Jossed for this film. Bebop and Rocksteady end the film as allies to the Turtles, though they may commit a Face–Heel Turn when Shredder debuts in a future instalment.

Leatherhead will have a Cross-Dressing Voices.
With the recent confirmation that Rose Byrne will be voicing Leatherhead in this film, it would mean that Leatherhead would have a Gender Flip in this adaptation, although there is a possibility that the character will still be male, as Leatherhead may have a Cross-Dressing Voices, they may have already picked Byrne to do a Cajun-accented voice for Leatherhead. Also as Kraang Prime from Teenage Mutant Ninja Turtles (2012), Kraang Prime was a male character but he was voiced by different female actresses, so it may be the same case of Leatherhead. In addition, the leaked Mutant Mayhem toys show that Leatherhead's leaked appearance still looks masculine.
  • Apparently, the toys use they/them pronouns for Leatherhead, indicating that they're non-binary in this version.

Leatherhead will have an Adaptational Villainy.
The leaked appearance of Leatherhead seems to indicate that he will look the same as his appearance from his first version of Teenage Mutant Ninja Turtles (1987), so everything seems to indicate that Leatherhead will be based on the 80s Leatherhead and not on his version of 2003 and 2012. Leatherhead in Mutant Mayhem will be a villain and enemy of the Turtles and not an ally as in the various adaptations of the franchise.

Possible Villains who could be the Big Bad of the film
Since it is pretty likely that Shredder won't become the main antagonist of the film unlike most others, it is possible that a villain could take his place until a possible sequel comes in the future.

Potential New Characters
With their being possibilities of their having new characters in the film, there are potential possibilities on new characters.

Potential Mutant Heroes
With a few mutant characters from TMNT confirmed, it is likely that the Turtles will form a team with them to stop the evil mutants.
  • Mondo Gecko
  • Ghengis Frog
  • Wingnut
  • Ray Fillet

Potential Mutant Villains
With it being unofficial mutant villains from TMNT confirmed, it is likely that there will be a group of evil mutants.
  • Leatherhead
  • Bebop and Rocksteady
  • Cynthia Ultrom

Baxter Stockman will be the Mayor or a candidate.
  • Just like the IDW comics right now.
  • Jossed. He is merely a scientist.

Baxter Stockman and Superfly will have a relationship like Jekyll & Hyde.
Maybe Stockman and Superfly are separate characters in TMNT: Mutant Mayhem, but maybe they don't want to reveal that maybe Baxter Stockman and Superfly are actually the same character but with Split Personalities, where Baxter Stockman would be like Bruce Banner and Superfly would be the Hulk and the relationship of the two would be like Jekyll & Hyde.
  • Jossed. Superfly calls Baxter his father.

There will be a Early-Bird Cameo of Shredder.
At the moment, Shredder will not be the Big Bad in this movie, but that does not mean that we do not see a reference or mention of Oroku Saki. Maybe Splinter will mention the Shredder for the only time, but he would prefer not to talk about Saki anymore, since Saki is a man nobody should mess with. So we would see a Flashback of Oroku Saki's past with Hamato Yoshi and Tang Shen. Where perhaps, Yoshi and Saki were very good friends in their Hamato and Foot Clans, but something made the two men hate each other and caused Hamato Yoshi to be expelled from his Clan and with that he was exiled from Japan to New York, where in the sewers he would run into the baby turtles and he and the turtles would come into contact with the Mutagen and thus Yoshi would mutate and become the mutant rat named Splinter.

Wingnut will have a Cross-Dressing Voices.
Since Natasia Demetriou has been confirmed as Wingnut's voice, it's likely that he could have a high-pitched tone, much like the version in Teenage Mutant Ninja Turtles: Shredder's Revenge.

Possible Characters that can be introduced in the film
Since the film will have a new character (assumed villain) named Cynthia Ultrom, it is possible that the film will introduced new characters introduced in the film.

  • A New Companion ally to The Turtles who may help them in a sequel film.

Donnie's blood will end up being a plot point
  • A scientist, possibly Baxter Stockman finds a sample of Donnie's blood from the sai wound and reverse engineers mutagen from it.
  • Jossed for Donnie specifically, but TCRI do try to take all four Turtles' blood when they brief capture them.

Bebop will have a Race Lift.
  • With the confirmation that Seth Rogen will not only be an Executive Producer, but will also voice Bebop, this may suggest that in this adaptation; Bebop will have a Race Lift, and instead of being depicted as an African-American man like in the other adaptations, here he will possibly be a Caucasian man.
    • Jossed (sort of), the official trailer hints that Bebop and even Rocksteady aren’t human in this adaptation, but rather were born/created as mutated animals.

Karai will have an Adaptational Backstory Change and Adaptation Species Change in TMNT: Mutant Mayhem.
  • For this reimagining of Seth Rogen from the TMNT franchise; Karai for the new movie is that she was not human before, actually she was just an ordinary snake (Karai was the pet snake of one of the scientists of the T.C.R.I. scientific corporation) that was mutated and was adopted by the Shredder.

The Foot Clan appears in a Post-Credits scene for a possible sequel.
In addition, the possible members that we could see would be:

  • Shredder
    • Confirmed
  • Karai
  • Tatsu

Other ideas for The Stinger
  • They parody all those other movies that open up the multiverse, with a surprise visitor seeking the Turtles' help - Usagi Yojimbo.
    • A mid-credits scene features Cynthia Utrom hiring the Shredder to capture the Turtles.

Superfly's gang of mutants will become the Mighty Mutanimals
By the climax of the film, Mondo Gecko, Leatherhead, Genghis Frog, Ray Fillet, and Wingnut will realize humans aren't all that bad and team up with the Turtles to stop Superfly, leading to them becoming a B-team to the TMNT for a potential sequel. Meanwhile, Bebop and Rocksteady will evade capture by the police only to find themselves confronted by the Shredder.
  • Confirmed, Superfly's gang, including Bebop and Rocksteady, pull a Heel–Face Turn and start living with the Turtles.

Superfly and Baxter Stockman aren't the same person, but are related to each other in some way
  • Superfly is the result of Baxter trying to cure his son of something.
    • Or he could be Barney Stockman holding a grudge against his brother, blaming him for his mutation.
    • Confirmed by this clip. Superfly calls Stockman his father. The film itself confirms them being adopted father and son in the opening scene.

The film will receive a PG-13 rating
Sadly jossed.

Leatherhead was once a human being.
  • Due to the fact that Leatherhead's right black boot is torn and their pants have damage to the hole where their tail sticks out, perhaps Leatherhead, before turning into a mutant alligator, was an ordinary male human who lived as a hunter in the Florida swamps. After transforming into a mutant alligator, Leatherhead's voice would change and become female, causing they to decide to become a non-binary alligator.

Splinter will claim to have been Yoshi's pet, then reveal that he is Yoshi in the sequel when the Shredder comes.
  • Jossed. The film confirms Splinter was just an ordinary, wild rat and never had an owner.

Superfly and/or Baxter Stockman will be revealed to be a Well-Intentioned Extremist .
  • And part of the reason why he wants mutants to rule the earth is to strengthen and better prepare the planet for an upcoming invasion by Cynthia Utrom (if she is indeed working for or in fact is Krang).
    • Sort of jossed. Baxter wanted to create a mutant family of his own and was killed for it, and now Superfly wants to avenge his dad.

     Post-Release 
Superfly is genetically Baxter's son
  • Unlike the other mutants, Superfly has hair resembling Stockman's, implying that Stockman may have combined his own DNA with Superfly's in some way, but didn't for the others or the mutagen itself. Alternatively, he used his own DNA to create the mutagen to cause animals to gain human attributes, which would also explain Splinter's afro when he was younger.

The main TCRI agent is this continuity’s incarnation of Agent Bishop

Alternatively, the main TCRI agent is Hun
Mainly due to his imposing build and the scars on his face. Maybe like in the IDW continuity, he's Casey's father.

Hamato Yoshi does exist in this continuity
However, rather than being Splinter's owner or Splinter himself, Yoshi was a martial artist who worked at TCRI to pay the bills and worked alongside Baxter Stockman on his mutagen. So technically, he's still connected to Splinter and the Turtles in a way.

     Sequel Theories 
Shredder, and by extension the foot, are an assassination-for-hire group.
  • They carry out orders that they are contracted for, but are otherwise independent of TCRI's operations.

Stockman will be revealed to be alive
  • But he will have been reduced to a head in a jar like the 2003 show.
  • Furthermore, we do see two TCRI agents dragging him away.

Jennika from the IDW comics will appear
  • She'll be a low ranking Foot member who Heel-Face turns and joins the brothers, and mutates into a turtle herself either from exposure to the blood TCRI already has or when one of the turtles is forced to give her a transfusion of their own blood.

Shredder will be Ch'rell from the 2003 series

  • Given that Splinter isn't Hamato Yoshi and has no real connection to him, it might not make a lot of sense to use the Foot Clan and Oroku Saki in their traditional roles. Having Ch'rell be Shredder could be an easier way to tie the character into Cynthia Utrom.

The Shredder is the real reason why Splinter hated humans
Just like in most continuities, Splinter was Hamato Yoshi's pet rat. Yoshi learned TCRI's secrets, and they hired Shredder to bring him to them so they could experiment on him. This will be a Cerebus Retcon explaining how Splinter knew about the milking machine. Splinter lied about why he hated humans and why he learned Ninjutsu because he didn't want to groom the turtles into being instruments for revenge. The Stinger for the next movie will then reveal Hamato Yoshi is still alive and is this universe's version of the Rat King.

The Shredder will be woman in this version
The Turtles will assume that their new (and likely greatest) enemy is a man, only to for them to be revealed to be female under the armor. At which point Shredder would state that it'd be weird if she was a man since her name is Saki, a joke about how the Shredder has had a feminine name in Japanese all these years.
  • It'll turn out that this version of the Shredder is actually Karai.
  • A Female Saki made sense since Saki is a woman's name or just give male Shredder an actual man's name to give some realism

The Shredder will be played straight as a terrifying threat.
The Shredder's presence will be constantly dominative and intense, with him being able to beat all the turtles without any trouble. It will serve as a major juxtaposition to the sequel's likely heavy comedic tone. They can do the tradition of Shredder mercilessly beating Leonardo one-on-one as a warning to the rest of the heroes.

And that's if Cynthia Utrom was a Canon Character All Along, but Maya Rudolph was Cast as a Mask.

The movie will explain how Splinter got his hands on traditional ninja weaponry
There's 15 years in between Splinter finding the turtles and the setting of the film, so Splinter might have went exploring throughout NYC and found a human Japanese immigrant who doubled as a ninja historian.
  • That immigrant also happened to be Hamato Yoshi.

Superfly's siblings will be Put on a Bus in the sequel
Or a case of Negative Continuity. The siblings will be nowhere to be found, and Splinter will be single again with no explanation given.
  • Perhaps if Put on a Bus, they'll be established to be doing their own thing, maybe even setting up a Mighty Mutanimals spinoff.
  • Bonus points if they do leave on a jerry-rigged subway train.

Hamato Yoshi will appear as an ally of the Turtles and Splinter in the sequel
Given they had Splinter's ties with Hamato Yoshi were Adapted Out, perhaps a sequel could take advantage of that by having Yoshi be an ally to the turtles and Splinter. Training them in ninjitsu and other martial arts to help fight the Shredder. Maybe he ends up being a Cool Uncle to the turtles as well.

Or... Hamato Yoshi IS the Shredder
He already killed Oroku Saki himself, and went on down the slippery slope after that.

Due to Bebop and Rocksteady undergoing Adaptational Heroism, Shredder will have a different pair of mutant henchmen.
Assuming Bebop and Rocksteady don't do a Face–Heel Turn, Shredder will end up using some ooze to create some mutants of his own to help with the Turtles capture. They could even act as Evil Counterparts to Bebop and Rocksteady. As for who could be in the role...

Splinter and the turtles are reincarnations of Hamato Yoshi and his sons, like in the IDW comics
The main difference is that not a single one of them know it at this point in time.

The Shredder will be a case of Beware the Silly Ones.
When he first appears, the Shredder will be shown to have a personality similar to the '87 Shredder, being very goofy and childish with much of his antics being played for comedy. The turtle will initially assume that he's no threat at all, but when they go to fight him, he'll be revealed to be an extremely competent, deadly fighter and easily mop the floor with them.

Hamato Yoshi will be revealed as one of the characters featured in the self-defense video Splinter and the Turtles learned ninjutsu from.
After the heroes get a beating from the Shredder, they'll ask him to train them.

Paramount Animation will have involvement producing the sequel.
In a similar manner to how The Angry Birds Movie 2 was co-produced by Sony's in-house animation studio. Paramount's in-house animation studio could have involvement as well.

Shredder will become a Dragon Ascendant.
Once he's faced the mutants and seen the miracle of the ooze, the Shredder will murder Utrom and his Foot Clan will take over TCRI to use its technology for his own ends.
