I don't mean "based on superheroes, but with the key markers of the genre filed off (like Smallville or Heroes)". I mean, it's about a guy in a colorful costume who goes out and fights crime on a regular basis.
If you discount short-lived flops like The Cape, the last time such a show was successful on American prime-time TV ... I think it'd be that Lois & Clark show from back in the 90's.

I think the creators claimed it was meant to be ambiguous or a different universe from the start but if you look at everything (Crisis changing Superman's family into the setup seen in the show, Diggle's first appearance) it really seems like it was meant to be main-universe at first. They use the separate universe for storytelling potential, but it is pretty awkward at points; I wonder if they separated it from a branding perspective because they want it to continue longer and the Arrowverse as a whole seems to be finishing.
Yeah, it was definitely sold as an Arrowverse show at first. There was a whole explanation in-universe about how the reason Superman wasn't around to help Supergirl with the crazy stuff she was going through in her current season is because he was leaving to do the things that set the groundwork for Superman and Lois, and there were a lot of intended ties between them at first.
Much like Agents of Shield over at Marvel, however, eventually the show separated from that idea and became its own distinct thing.

Gotham Knights on the other hand is almost certainly its own universe.
