Leo Vincey is charged by his dying uncle to follow the route in an old journal left by their ancestor John Vincey, who set out five centuries earlier to find the fountain of youth. His expedition (which also includes Horace Holly, a guide named Tugmore, and Tugmore's daughter Tanya) discovers the ancient lost city of Kor, ruled by the immortal She Who Must Be Obeyed, who loved John Vincey and believes Leo to be his reincarnation.

* AdaptationalAttractiveness: Horace Holly is described in the novel as being hopelessly ugly, even ape-like in appearance. In the 1935 film, he's played by Nigel Bruce, who might have let himself go a bit since his youth and is hampered with a terrible false moustache but is still far from deserving that description.
* AdvancedAncientAcropolis: The lost city of Kor, where She Who Must Be Obeyed rules.
* BettyAndVeronica: Leo has to choose between the cold but fascinating She Who Must Be Obeyed or the more normal mortal Tanya.
* IdenticalGrandson: Randolph Scott plays both Leo Vincey and the ancestor, John Vincey, whom She believes Leo is the reincarnation of.
* MysteriousAntarctica: Albeit on the North Pole, but for the same reason. The film changes the setting from Africa to the Arctic, at the time still mostly unknown and thus viewed like DarkestAfrica was in its time.
