- Acting for Two: Mark is the only person among the cast to play more than one character — the Mayor, the Colonel, and Markiplier. This is significant because Damien and William eventually become one half of Darkiplier and Wilford Warfstache respectively, both of whom are Mark’s alter egos.
- Blooper: The series is ostensibly set in the 1920s, but the dates on Little Buddy’s camera feed shows it to be taking place in 2017. Mark later admits in a livestream that he forgot to edit that part out.
- Creator's Favorite Episode: Mark stated in his "I EXPLAIN EVERYTHING" stream that his favorite episode of Who Killed Markiplier? was 3.

- Mick's (The Detective) line "Look. You're a real smart son of a bitch. Handsome, too. Beautiful, even." was supposed to be directed towards the viewer, but he accidentally said it to Tyler (The Butler). Mark and co. liked it so they decided to keep it in the final product.
- In Wilford 'MOTHERLOVING' Warfstache, the scene at the beginning where Abe has trouble pinning a knife to the Colonel's photo was completely accidental, it was kept in because it fit the episode.
