Bump: I've only seen that one page of the webcomic with the question and answer, so I don't know anything else about the plot. I'm pretty sure the webcomic page is linked and listed as an example of some TV Trope somewhere, but the first trope I checked was Gender Bender and it's not listed as an example there.
If not for this anchor I'd be dancing between the stars. At least I can try to write better vampire stories than Twilight. The Man, I Feel Like a Woman page has a quote from It's Walky! which looks like the one you're after. The comic's search function finds the specific strip, I think.
Interesting, so the webcomic I'm thinking of isn't the only one to draw that comparison. The webcomic I'm thinking of was in color, the panels were arranged vertically, and the characters definitely didn't have a Suggestive Collision right before the question. (And before anybody asks, no I didn't forget the Suggestive Collision, and no I wasn't looking at the same page on a different monitor in a different format.)
On the other hand, this page is arguably funnier because of the Suggestive Collision, and I specifically wanted to show somebody this gag. Mission accomplished anyway, thanks for your help!

(Not El Goonish Shive) A lady got transformed into a guy. Someone who knew what happened asked the transformed character, "What's it like having male genitalia?" and the first comparison drawn was "It's like having a little dog that barks at everything."
