I think the joke is lost on anyone unfamiliar with the character and/or pronunciation script.
The Revolution Will Not Be TropeableThere is a "t" in there, and you can tell she's stressing over a pronunciation. All you have to do is recognize it as the phonetic alphabet, and pretty much everybody has looked up something in the dictionary.
This trope is difficult to visually illustrate, and since the IPA is really the most universally-accepted method of transcribing sounds with a higher degree of accuracy, I think it should work.

"Peridot" is pronounced PEYR-uh-DOT, not PEYR-uh-DOH, as you might expect from other words that end in -ot, by the way. Hard T. Which is what that image is saying.

The implication is that the character's name 'Peridot' is pronounced Pari-doh, and she's stressed because the shadowy figure is pronouncing her name wrong. (As opposed to simply because a shadowy figure is calling out for her as might be suspected in the absence of the phonetic alphabet panel.)

There's a good chance it might fly completely over a non-native speaker's head, since as far as I've seen, English classes hardly ever touch the subject of "Those funny simbols in the dictionary". In my case, even after learning what those symbols are, I still don't know what pronunciation each one represents.

It's the International Phonetic Alphabet. See all those little symbols?
Granted, a lot of languages don't need them, because pronunciation works in an Exactly What It Says on the Tin fashion, but the IPA is not limited to English.
