It has to be within a certain proximity of the girl or else her body loses its psychic connection to the gem and she passes into a comatose state until the Gem is returned. As a phylactery, it is literally the housing container for her soul and must be near her at all times.
Do you see the ornate flower on the side of her head? That's the spot. Hit that with a bullet and she dies instantly. All other damage is fleeting.
That is true. Madoka magical girls have a sort of MP system where their soul gems degrade with every use of magic and have to be refreshed with Grief Seeds obtained from killing Witches. Their emotional state also affects the degradation which, in turn, seems to affect their emotional state.
If the Gem degrades too far, the magical girl succumbs to despair and transforms into an incredibly powerful reality-warping monstrosity that remorselessly murders anyone in their vicinity with seemingly little rhyme or reason.

My Tumblr. Currently liveblogging Haruhi Suzumiya and revisiting Danganronpa V3.As part of this discussion of... Mami vs Vash? So weird. Anyway, I just want to point out that once Mami believes something is right she'll set her mind to do it and she'll do it quickly.
A very important scene to point out is when she found out in the second timeline (iirc) that magical girls would become witches. Whereas every other magical girl in the series were tainted by despair and reacted in horror, Mami immediately sought to solve the problem and chose the optimal route. She was able to restrict Homura's movement and abilities before she had time to react to it (knowing that Homura would use her powers to stop her) and quickly aimed at taking down the rest of the magical girls in order to prevent them from becoming witches (since she was well aware that they'd only end up hurting more people in the end).
Point is that Mami showed split-second decision making capabilities and clarity of thinking even in dangerous and stressful situations. Combined with her ability to manipulate the ribbons to restrict movement, her gun creation abilities, and the fact that she's virtually invulnerable unless someone specifically targets her soul gem... i think she might be able to defeat Vash. Keep in mind that Vash has no reason to believe that shattering some gem on her clothing will kill her. For that matter neither does she, so it's not like Vash can figure it out because she's being weird about the gem to protect it.
It's not even that convoluted a death battle to make either; not only is Vash a super wanted criminal, but he's shown the capability to destroy entire cities (and when that power is unleashed he doesn't have much control of it either, iirc). Mami would totally be willing to target him. (An easy scenario to imagine is that Mami is made aware of some super dangerous being in a nearby city, and when she gets to it it's completely in ruins except for Vash)
About that... Immediately after finding out that magical girls become witches, Mami's soul gem remained completely clear... in fact I don't think her soul gem ever shows signs of corruption. So I don't think that's a viable means for Vash to win the fight... er, lose the fight... or both? Either way it wouldn't really apply to Mami.

So are we just ignoring that Vash can channel his Plant powers into his gun to fire bullets that can send matter into subspace, and a second type that can erupt into miniature black holes that consume everything in a 1 km radius, or the fact that an uncharged blast from his Angel Arm could reach the Moon from the surface of the planet in seconds, and create a massive crater in it the size of a small country at least?
Hell, Vash should be as fast as his brother Knives, who could casually reach the stratosphere from the surface, in a few seconds at most.

I would consider a more accurate reading of that scene to be that Mami suffered a complete emotional breakdown and started murdering her friends. You make it sound like this was a cold and calculated decision she made, as opposed to the belligerent scream-cry-ranting she was doing in the scene in question. It is very likely that she would have become a witch shortly after this scene had she not been killed on the spot during the attempt.
This scene was many things, but a demonstration of Mami having incredible emotional fortitude and calculating nature is not one of them.

Yes. Mami either exhausting so much magic that her mana runs dry or being driven to emotional despair over the course of the fight is actually a very bad thing for her adversary, because it triggers a permanent transformation that makes her significantly more dangerous.

If they reveal more for Ichigo in his preview in comparison, it makes me believe he'll be more likely to lose instead.
Watch SymphogearMami did pretty much match Homura in firepower with single shot weapons while Homura was using multiple automatic weapons. She also tricked Homura with a clone that was made of her ribbons. In multiple timelines, it does figure out how to fight Homura.
While she certainly snapped in that mentioned timeline she did have a pretty well thought out plan on how to kill her comrades. The advantage for Vash is witch Mami is probably not as deadly as Madoka witch who can destroy an entire planet

On the other hand, he's one of those characters that either wins most of the time or gets curbstomped by much stronger characters.
EOS Naruto is in range with EOS Vash, actually, and both are about Saiyan saga level dragon ball.

Well let me not oversell it - a lot of his characters have some twisted mindsets, depressing conclusions to their story arcs, or just come from some dark places. Yu Yu Hakusho frequently involves Body Horror, Humans Are the Real Monsters, Uncanny Valley, And I Must Scream in it's characters and their fates. It's not like Berserk or Warhammer 40000 where everything is part of a Crapsack World, but minus the gratuitous rape and sense of utter hopelessness,
I'd like to say this video about one of Yu Yu Hakusho's main antagonists sums up what I mean. The complexity and twistedness of many of his characters (or at least the villains) makes me less surprised that he would come up with something as eerie as Hisoka.
But then again, visually, Clowns Are Just Creepy.
There's also the possibility it's just a Running Gag of some kind - there was a clown character in YYH as well.

Although, I wouldn't be surprised if that's because there isn't a 15 second tangent from Boomstick.

If not for the fact that Dante already had his match, I'd say that a better choice would be Vash vs Dante.

One Strip! One Strip!I can see that but I sort of felt that was bit to on the nose. Nero and Vash I felt were an interesting match in that both are gunslingers that have been voiced by Johnny Yong Bosch but with different personalities and power origins.

I think in JLA/A, Aquaman hits Namor with a whale. Also his communicate with fishes has been demonstrated to be able to give Namor a headache.
