In general, Peter's life on 6160 comes across as a huge Be Careful What You Wish For reality. In the original comics, Peter famously grappled with how much being Spider-Man seemed to ruin his life, and has contemplated abandoning the mask many times and has wished he was never bitten by the spider as a result. He basically got his wish in this universe and possibly more: his parents were around to raise him, Uncle Ben is alive, he's married to Mary Jane and has children with her, he has a stable career with the Daily Bugle where J. Jonah Jameson is actually nice to him, and most importantly, he was never bitten by the radioactive spider and was never burdened by being Spider-Man. And yet... Peter feels a very pervasive emptiness in his life that the Maker's interference with his destiny caused. For all its ups and downs, Peter being Spider-Man was good for him as well as every single life he's saved, and the Maker had robbed him of all sense of purpose by preventing the spider from biting him, and thus robbed this Marvel Universe of one of its greatest champions.
Peter: It's just... for the longest time, I've thought something was wrong with my life, Ben— that something was wrong with me. I can't really explain it, but now I just... I just know that I've been right all along... And I don't know what to do.
Peter's parents die when he was fifteen instead of when he was very young. While this means that he gets to know his parents more than his 616 counterpart did, this also brings up the possibility that their deaths would hurt more due to Peter's inevitable closer bond to them since they were around to raise him this time.
MJ notes that she's never seen Ben cry, which makes it even more poignant that he's shedding Manly Tears at the memorial service for May and other victims of the Illuminati's terrorist attack.
