At one point in "Whatever Happened to Shorty Twang?", Sylvester accidentally hurts Tweety. He is quick to sincerely ask Tweety Bird if he's alright.
The Grand Finale "This is the End" has Sylvester appear to finally eat Tweety Bird, eventually resulting in the show getting cancelled. After it's revealed that the episode's events were All Just a Dream, the first thing Sylvester does upon finding out that Tweety is still alive is to hug him and tell him how much he loves him.

Usually, when something happens to Sylvester, Granny just dismisses it as the cat fooling around, but in the climax of "El Dia de los Pussygatos", upon seeing a skeleton chase after Sylvester (who turns out to be the missing mayor, Abel Valenzuela), Granny is shocked and chases after the skeleton. Tweety even helps by setting up a wall in an alley for Sylvester and the skeleton to crash into.
