Hawke's Bay

  • While out on the balcony, Alma tells Orson that he’ll need to be present at an exchange with Kronstadt Industries in her place, as she’s too valuable to lose if it happens to be a trap. He’s initially upset, but accepts it, saying he just wants Alma to get safe. It's nice to see that despite all his whining and all the jabs Alma takes at him, Orson still cares about her.
  • The titular "Nightcall": After sleeping for an hour, Alma wakes up and calls her sister Celia. It's revealed that Alma had a daughter with Sean Rose. The girl's name is Mercedes and she’s living with Celia. Alma was calling to apologise for missing Mercedes's birthday, and laughs proudly when she learns that Mercedes got a part in her school play. The call shows a sweet, motherly side to Alma Reynard, far from the Faux Affably Evil terrorist she was initially presented as. Overlaps with Tearjerker, as you still have to assassinate Alma before you leave the mission.
    • The fact that Mercedes seems to be living a regular life with her aunt, with birthday parties and school plays, far from the hectic, bloody and ultimately ill-fated lives her parents led.
    • Possible Fridge Brilliance: Mercedes's name means and can be shortened to "Mercy". It might be ironic a merciless terrorist like Alma Reynard named her daughter after the virtue Mercy, but maybe that's the point.

Miami

  • While normal Hitman targets are usually seen as impossibly bad people, Sierra Knox comes off as a fully fleshed out person who runs the gambit of emotions. She's staunchly protective of her father, despite their shaky relationship, and will literally kill for him (in fairness, the person in question was blackmailing her father). She also has a Friendly Rivalry with Moses Lee, her competitor, and the two have fun drinking games. 47 can play this game if disguised as Lee. If it plays through and he wins, she will be a Graceful Loser and look forward to the next race.

Santa Fortuna

  • The "Hero of Santa Fortuna" challenge, while very tedious to get through, leads to several heartwarming moments where you experience gratitude from both Delgados and Andrea Martinez for a job well done.
  • When Andrea actually does give in and read the letter from Hector while on the balcony, she actually monologues about the good times the two spent together and it's clear that she still cares for him as much as he does. She just thinks them being together would prove too dangerous for her.

Whittleton Creek

  • 47 never has much interaction with people short of killing them, but in Whittleton Creek, he can help the neighborhood. He can help the aged and struggling mailman with some parcels and find some propane so that the family barbecue can get started. The overflowing gratitude of the people feels authentic.
    • This is sort of mitigated in the mailman's case, however, as listening to him once he's finished with his rounds will reveal that he actually hates the residents of the suburb.

Isle of Sgàil

  • Janus's wake is a supremely touching moment as the members of the Ark Society pay their final respects. Some of them tell Janus that they wish they knew him better, lament they never finished their chess game and commend him on a life well lived. Others compliment the eccentric and lonely mortician for making Janus's body seem so lifelike. The funeral director and Constant take the cake: She speaks like an old flame saying farewell to her lover, and the Constant vows to finish Janus's work. Only Zoe ruins it, whispering taunts into Janus's ear.
  • The Slap-Slap-Kiss relationship of Sophia and Blake Nathaniel. A dialogue between Blake and Zoe has the two of them discuss the necklace. While Blake is worried Sophia might try to steal it, Zoe points out that Sophia doesn't want that. What she wants is for Blake to put it around her neck. 47 can use this as an assassination attempt, but for just a split second before he strangles her, Sophia has a serene smile around her neck as she admires herself.
