Tsukune Aono, a human teenager, was enrolled into a school called Yokai Academy. When he arrived, to his horror however, he discovered that his new school was actually populated by real monsters who don't take too kindly about humans.
He tried to escape but after a incident with one of the students, Tsukune, along with his new friend, Moka Akashiya learnt that he is able to use abilities akin to that of a spider.

- He Knows Too Much: Warren orders Patton to kill everyone but Tsukune for finding out about his plans to eliminate humanity.
- Humans Are the Real Monsters: Many from the Monster World feels this way about humanity, especially due to their role in murdering the entire Spider Clan, the ones that helped form a truce between both sides,
- In Spite of a Nail: Moka, Kurumu and Yukari still deal with Ginei despite Tsukune being at the human world.
- La Résistance: There are monsters like the Bus Driver are attempting to prevent Warren's plans to succeed. Tsukune and his friends would join as well.
- Mythology Gag: Tsukune used a makeshift suit made from junk (e.g. red sneakers, sleeveless blue hoodie, red ski mask etc) looking like a version of the first Scarlet Spider suit.
- Oh, Crap!: Yukari discovers that the Blood Moon will occur in six months.
- Off with His Head!: Monster Tsukune rips into Patton's neck and rips off his entire head.
- One-Winged Angel: Both Tsukune and Patton turned into monstrous spider forms at the end of Season 1.
- Power Limiter: Moka's rosary is what keeps her powers under control. Moka would give it to Tsukune to keep his monster form dormant.
- The Reveal: Part 5 has some major revelations.
- Tsukune is actually a Half-Human Hybrid born from a human and a spider monster. His powers were actually dormant until Moka came and bite him, triggering them.
- Headmaster Warren was the one that created the spider monster that would become one of Tsukune's parents.
- Yokai Academy's true purpose is to transform students into Manchurian Agents that will infiltrate the human world and start attacking them when the time is right.
- The Bus Driver is the one that invited Tsukune into Yokai Academy in hopes he could stop Warren's plans.
- Running Gag: Everyone keep pointing out that Moka's bite might be the reason Tsukune turn into a monster with Moka being very vocal that isn't the case. This actually is a major plot point as Warren reveals that Moka biting him did actually awaken the dormant spider powers within Tsukune.
- Too Good for This Sinful Earth: The Spider Clan were benevolent monsters that tried to stop the war between Yokai and humans, even offering one of their own to be part of a marriage arrangement. Even though their actions brought about peace for a while, a few humans chooses to exterminate them.
- Wham Line: When Tsukune returns to his room, he bumps into a scrawny boy with glasses. Both boys introduces themselves.Tsukune: Name's Tsukune. Tsukune Aono.Student: My name is Patton. Patton Parnell.
- Wham Shot: Outer Moka bites onto Tsukune’s arm which reveals that his blood is now....purple.
