The only way for the host to rid themselves of this is to either transfer the Mark to someone else (like Cain did to Dean) or destroy the Mark entirely (which if you read the rest of the tropes on this page [[{{Understatement}} is a very bad idea]].
* DangerouslyGenreSavvy: The reason that Death wants Dean to kill Sam is because he knows as long as the latter is alive he will never stop trying to find away to free Dean from the Mark.

And while it hasn't been stated yet murdering Death is most likely going to have far reaching negative consequences as well.
