** In the series, [[FatBastard Takiji Kuroido]] was, while not a major player in the main conflict, a recurring background antagonist with strong relevance in the backstory. Here, he makes one appearance, has no dialogue, and gets a hole punched through him by Nui in an almost forgettable moment. Though given the point in the timeline [[ForWantOfANail at which the changes start]], he still carried out the first assassination attempt that resulted in Soichiro changing his identity.

* AdaptationalVillainy: Isshin in canon, despite his more unscrupulous aspects, is treated as the posthumous BigGood of the series. This Isshin, thanks to being alive, starts out the same way but begins to show more of a willingness to sacrifice others to achieve his goal of rebelling against Ragyo, actively manipulating Ryuko while enabling Satsuki to oppress the students of Honnouji Academy, pitting the two against each other, and allowing spies to fester in Nudist Beach. The biggest moment of this is when, in Chapter 23, it is revealed that [[spoiler:he drugged the Mankanshoku family, experimented on Mako with Life Fibers, and ''knowingly'' got Barazo, Sukuyo, and Guts killed to save himself]].

Only Mako continues to serve up oblivious humor, and even then, Mataro's death and Ryuko's failure to prevent it do away with that for many chapters. Six months before Ryuko's journey begins, Nui Harime accidentally goes to the wrong address in her mission to kill Isshin Matoi, resulting in the deaths of Mako's parents and far-reaching shakeups for the entire cast.

cast. Since Isshin Matoi is alive, he is free to continue leading the Nudist Beach rebellion against Ragyo Kiryuin and REVOCS. When Ryuko comes to his house, he introduces her to Senketsu, the friendly Life Fiber, and makes her a member of Nudist Beach, training her in the ways of the organization and their mission. Eventually, he sends her to Honnouji Academy to fight Satsuki Kiryuin- and now that she has been trained, she is able to overcome her opponents much easier than she did in the original show.

Meanwhile, Mako and Mataro have been left with severe scars from their encounter with Nui. Mataro, in particular, dedicates himself to growing strong enough to avenge his parents. Mako still befriends Ryuko, but has a lot of baggage underneath her cheery attitude. And with Nui on their trail, things are bound to be mixed up even more...
