Does the Founder of the Kingdom have to be dead/missing by the time the story proper begins, or can he be a contemporary of the story's main cast (i.e. the nation was rather recently founded), if not one of them in the first place?

He doesn't have to be dead, though the trope will be played very differently if he's part of the main cast.
