Generally, the main problem right now that I see from playing the game for about 2 hours (Dota 2) is the matchmaking process. It takes forever, literally, you get 9/10 people connecting to the game, then the 10th person declines to join, placing you back into matchmaking for another 10 mins until you actually get a game.
Also, another problem which would end up hurting them is the fact that Ice Frog wont want to change the game so much from the original Do TA, so it'll essentially be the exact same game, except with matchmaking, VA, and better graphics and models.
The game itself also isn't very friendly to new players, seeing as how Do TA from WCIII wasn't very new player friendly either. Perhaps a matchmaking system like Lo L's or similar would help the newer players and maybe even a tutorial could help the newer players get a hang of the game.
Also another thing: the bashing is kind of worse now that people can actually use mics in game without the use of vent or anything.

If perhaps only certain players were nominated to coach new players then perhaps it could work. Although many people have different play styles and the play styles of the coaches might not be the best for newer players.
Also something maybe they should consider for Do TA 2 is showing the range of which the spell can be casted, such as in a circle showing the range of the spell (i know they have a cir le for target Ao E spells) this was done in Lo L and it seemed pretty helpful for the newer players

Anyway, I got my invite like 2 weeks back. As a complete Dota newbie, with some experience in League of Legends, I'm pretty happy that my IRL friends are pretty good at the game. I've been slowly picking it up, and my favourite heroes are the supporty ones with big nukes.
Favourite heroes at the moment are Ancient Apparition for his awesome skills and auto-attack animation and Kunkka for his awesome design.

The sheer volume of voice clips is amazing. I doubt I'll actually pay to play a sequel-copy to a game I was horrible at, but depending on the price I might buy this game just to support Valve.
