The producer once again seems to indicate that a crossover with the Flash is (unfortunately) unlikely, but he does say tonally, the show is more like Flash than Arrow.
Between this and some of the articles about The Flash, I'm beginning to like interviews where the interviewee answer questions in ways that don't actually field the questions asked. Tres vague.
That said, that joke about how both not only do both Barry and Supergirl share a tone, but they both died in Crisis on Infinite Earths at least might indicate that this Supergirl here is Kara Zor-El, if there was ever any doubt.
At this point I'm most curious about Superman itself, since his presence (or lack thereof if the case may be) will likely have a decent effect on how the setting works: is it going to be like how DC often does character focus series, with the "main" character of the mythos only occasionally appearing? Is Clark Kent going to be a recurring character, but not Superman himself? Could Superman be appearing in a mentor role? Or could it be none of the above, with Kara existing in a vacuum of sorts.

1 - He was never born
2 - He was born, but died on Krypton as a baby.
There was a description for the series that said "Since arriving on Earth, shes been hiding the powers she shares with her famous cousin Superman", which a lot of people have taken as confirmation that Superman is around and well known in this world, but i took it as nothing more than real world reference to them being cousins in the comics, not that he'd be around in the show.

Confirmation of no crossovers, at least "for now". The exec also says they've been shown the costume and it looks "awesome".
The Audition tapes reveal that Superman does exist but this show doesn't take place in Metropolis so Superman is only mentioned.
Kara lives in National City.

No Kara Zor-El didn't change her first name when she was adopted by the Danvers. Usually she is Linda Lee Danvers.

We know who is playing Supergirl, we know Superman exists and is famous, and we know it takes place in National City which is on the other side of the country.
National City is a reference to DC Comics original name National Allied Publishing but they were also called National Comics after merging with Detective Comics. They were officially National Comics until they changed their name to DC Comics in 1977.
Eventhough they were National Comics they still used DC Comics as a nickname.

Age Lift: Kara is usually in her teens when she is Supergirl but on the show she starts out at the age of 24.
Mythology Gag: The show takes place in National City which is a refence to DC Comics name they had at their founding.

Canon Foreigner: Kara has a foster sister Alex Danvers on the show while in the comics the Danvers never had any biological children.
Adaption Name Change: On Earth Kara Zor-El is going by Kara Danvers instead of Linda Lee or Linda Lee Danvers.

The chances of a horse named Comet showing up are actually pretty high, given how Mythology Gag heavy these series tend to be.
The chances of that horse actually being superpowered are likely lower than said horse's shoes, however.
Though, on the other hand, Post-Crisis Comet (Andrea Martinez) is a possibility, albeit likely with a much less convoluted backstory.
