  • The titular desk job is nothing more than inspecting Aperture-made toilets. Fortunately, the player only has to deal with the "boring" version of the desk job for the first three minutes of gameplay; things start to get more interesting from here.
  • The intro has Cave Johnson giving a rousing speech, calling the main character a genius and that their work will matter in the future. Then it turns out Grady put in the wrong tape, and what Cave Johnson actually says to rank-and-file employees (i.e., you) is much simpler:
    Cave Johnson: You. In the overalls. Get to work, or you're fired.
    Grady: They don't give out overalls, anymore.
  • Everything involving the invention of (this version of) the Aperture Science Turret. Especially since the prototype turrets seen here are made out of toilets.
    • And how did Grady come up with the idea of the turret? He and the player saw a toilet that was smoking, then Grady fell in the toilet when it blasted him with water, then the player's attempts at getting Grady out wound up breaking the Ammunition pipeline overhead…just in time for a ton of bullets to spill out of it and into the water bowl of the toilet. The comic timing of it has to be seen to be believed.
    • It takes six months for Grady to make the toilet turret. Well, it actually took him one month. The other five was for another toilet turret, with a whole bunch of different weapons glued on to it.
  • Somehow, the player still has their desk, even when in prison!
    • Just the idea that Aperture had its own maximum security prison facility is worth a chuckle, if only for how hilariously dystopian the idea is.
  • While depressing, Cave Johnson's ultimate fate is also darkly amusing. Especially when he explains why he's been all on his own, stuck in a giant head in his likeness:
    Cave Johnson: So the lab boys put me in the big head, switched me on and... uh... I don't know, I panicked. Killed a few of them. Said I'd kill more if they ever came back. Honestly, I-I regret that now.
    • And just when it looks like Grady and the player finally ended Johnson's suffering, the back-up power to his head comes on.
      Cave Johnson: [frustrated sigh] …You're both fired.
  • The Running Gag of in-between scenes where a group of mantis discover a light bulb and manage to BUILD A ENTIRE FREAKING FUTURISTIC CITY out of a simple light bulb. At least until the final act, with Cave's giant robot head having destroyed the city when it fell down.
