Is it true that Total Biscuit defended someone who organized DDOS attacks on Microsoft? I've not heard much about this.

Unless you're saying that the guest once organised DDOS attacks? Which is both something I haven't heard of and also doesn't actually factor into if the abuse she received after being on the show was in any way okay (hint, it was not okay).

I can't remember the whole story but is was alleged that someone at Microsoft "misgendered" Laura and afterward were attacked verbally and possibly DDOS'd by Laura's supporters, who were personally spearheaded by Laura into doing it. That was before Total Biscuit made the soundcloud commentary.
Again I'm not sure how true or false it is, as rumors are going around widely. DDOS attacks are not okay either.

It's likely that what your hearing is just a rumor.
I actually had a look at the reddit thread on the soundcloud. There is no mention of anything involving DDOS attacks, and by all indications, Laura didn't "spearhead" shit. It was other outlets reporting on it, and all she did was complain a bit on twitter, and then other outlets (like Kotaku) decided to run with it for clicks. Kotaku actually refused to take down the article when Laura asked them to.
Sooooooo yeah.

Fanfiction I hate.Wonderful, I find it funny how many journalists or media workers make themselves look like fools via just to trying to take out Total Biscuit in stupid ways....
It's quite clear they fear his influence....
It may have just been a rumor, then. Hopefully so. I don't think anyone could justify supporting DDOS attacks on innocent people.
I can see why some people hate him and some people love him. Some of his videos annoy me, some I like. I'm neither a "fanboy" nor a "hater" in that regard. It's one of few cases where I can see logic behind both spectrums.
He does have some very annoying fanboys and anti-fans alike. Some see him as God's own gift to humanity, others as some worthless piece of slime. Both equally weird.

That is the face of a man who just ate a kitten. Raw.The funny thing is he has made it very clear he hates both his crazy fanboys and some his equally obsessive anti-fans.
I am in the same boat. I like some of TB's stuff and I don't like other stuff that he made. His personal life other then the fact that he was at the threat of becoming very sick due to his now defunct cancer does not concern me.
His opinions don't influence me in any matter. They are just worth considering amongst others. It helps that he generally backs up his opinions though and I find him a critic worth respecting. I rather form my opinions then to follow the love or hate crowd

The general allegation was that he ignored the presence of DDOS attacks or at worse silently supported them, which is what some assumed he was doing there.

Like you haven't even sourced the existence of this as a rumour, never mind one with any basis in fact or one that TB has been told about.
Also what's with the quote marks around "misgendered" (before anyone asks, mine are there because I'm directly quoting)? You're not quoting anyone but put quote marks around the word...

It was a popular topic on 4chan's /v/ a little while ago. This was the first I heard about it however.
The same remark about Laura wasn't sourced and it wasn't clear what was meant by "misgendering" or who did it. Allegations range from DDOS attacks being thrown on Destructoid to Microsoft itself by Laura's fans for the reason "Misgendering" yet nothing was provided to back that up either.
Looking more into it it doesn't seem any evidence exists either way; no DDOS attacks from Laura and fans, no insults from Destructoid or other group.
I'm not sure why a handful of people are so defensive over me asking a question about an allegation I picked up on.
