So I have a villainous Four-Temperament Ensemble revolving around a Leukine mastermind.

Each of the four are literally parts of the leader's personality manifested. The reason for the quartet's creation is to stabilize the leader's thoughts and behavior so that he may channel his own powers much more efficiently.

In the course of my story, one by one each of the temperament embodiments are sealed inside their signature weapons or equipment. Each time having a negative effect on the Big Bad's mental health.

After the last temperament is sealed away, how should the Big Bad sound when he speaks?

I can either go with Mood-Swinger, Creepy Monotone, or both.

I'd say both. Start off creepy monotone, then follow up with some kind of epic spaz-out of volcanic proportions. At least that's what makes sense to me, in my mind insane villain = Large Ham.

Well if its sealing them away, I'd personally think the Creepy Monotone voice would be best. Unless the other temperaments could somehow leak from time to time to his consciousness (but just as quickly leave), I can't imagine him having much emotion at all. But that's just me.

Guess it depends whether they're literally externalised emotions, or just symbolic 'forms' to help keep the emotions, that remain behind, stable. Are the protagonists sealing the mold or the jello?

Now that you ask I might have to say the jello.

The most extreme traits of the temperaments is turned into a separate being.
